摘要
目的:通过网络药理学探讨中药牡丹皮治疗系统性红斑狼疮(SLE)的作用机制。方法:通过中药系统药理学数据库(TCMSP)筛选出牡丹皮活性成分及作用靶点,并在人类孟德尔遗传数据库等常用数据库筛选出SLE疾病靶点,应用R语言软件筛选牡丹皮治疗SLE的靶点,然后构建中药-成分-疾病-靶点调控网络,其次通过构建牡丹皮-SLE疾病靶点的蛋白互作网络(PPI)和条形图,筛选核心靶点,最后对其结果进行GO功能富集分析和KEGG通路富集分析,得出牡丹皮治疗SLE可能的作用机制。结果:通过筛选获得牡丹皮活性成分11个,得到牡丹皮治疗SLE可能靶点97个,通过筛选获得AKT1、VEGFA、MAPK1、IL6、JUN、CASP3和MAPK8等关键靶点,这些靶点主要涉及细胞对脂多糖的反应、对细菌起源的分子的反应、对氧化应激的反应等生物学过程,主要富集在PI3K-Akt信号通路、流体剪切应力和动脉粥样硬化等信号通路上。结论:牡丹皮通过多途径、多靶点、多通路影响SLE的发生。本研究初步揭示了牡丹皮治疗SLE所涉及的相关生物学过程及信号通路,为后续更进一步研究提供理论参考和依据。
Objective:To explore the mechanism for treatment of systemic lupus erythematosus(SLE)with Cortex Moutan through network pharmacology.Methods:TCMSP was used to screen the active ingredients and targets in Cortex Moutan.While the Online Mendelian Inheritance database and R language software were used to find the SLE disease and therapeutic targets.Then a traditional Chinese medicine-component-disease-target regulatory network was build.Through the Cortex Moutan-SLE target protein interaction network(PPI)and bar graph screening,the core targets and finally GO function enrichment analysis were performed on the KEGG pathway to obtain the possible mechanism for treatment of SLE.Results:11 active components of Cortex Moutan and 97 possible targets for treatment of SLE were obtained.Some key targets,such as AKT1,VEGFA,MAPK1 and IL6 etc.which targets mainly involved cell pairs were found.Biological processes such as the reaction of lipopolysaccharide,molecules of bacterial origin,and oxidative stress are mainly enriched in the PI3K-Akt signaling pathway,fluid shear stress,and atherosclerosis signaling pathways.Conclusion:Through multiple pathways and targets,Cortex Moutan affects the occurrence of SLE.This study initially revealed that the related biological processes and signaling pathways for treatment of SLE by Cortex Moutan,providing theoretical reference and basis for further research.
