摘要
为研究杂交构树青贮(Broussonetia papyrifera silage)对务川白山羊瘤胃微生物群落结构的影响,选择64只健康务川白山羊,随机分为4组,每组16只(公母各半,分开饲养),粗饲料分别饲喂100%青贮玉米(CS组)、20%杂交构树青贮+80%青贮玉米(LBP组)、50%杂交构树青贮+50%青贮玉米(MBP组)、100%杂交构树青贮(BPS组),预试期15 d,试验期172 d,试验结束每组随机抽取6只山羊(公母各3只),经口腔采集瘤胃液,采用16S rDNA测序分析瘤胃液菌群组成。结果显示,16S rDNA基因测序共获得6861个操作分类单元(OTUs),属于20个门230个属。BPS组的OTUs、Observed_species指数、Shannon指数和Chao1指数分别比CS组高20.99%、27.23%、9.73%和20.83%(P<0.05);门水平上,拟杆菌门和厚壁菌门是各组的优势菌群,BPS组拟杆菌门相对丰度分别比LBP组和MBP组低15.89%和20.17%(P<0.05);属水平上的优势菌群为普雷沃菌属、未分类拟杆菌属、未分类瘤胃球菌科;BPS组普雷沃菌属的相对丰度分别比CS组和MBP组低39.86%和39.25%(P<0.05)。综上所述,添加较高比例杂交构树青贮可显著提高务川白山羊瘤胃菌群丰富度,添加50%杂交构树青贮时瘤胃拟杆菌门和普雷沃菌属相对丰度较高,有利于促进瘤胃对蛋白质、淀粉和植物纤维的降解。
The aim of this experiment was to study the effect of hybrid Broussonetia papyrifera silage on rumen bacterial structure of Wuchuan white goat(one of Guizhou native goat breeds).A total of 64 Wuchuan white goats were divided into four groups with 16 goats per group(8 male goats and 8 female goats).Mixed forages were fed with 100%corn silage(CS group),20%hybrid Broussonetia papyrifera silage+80%corn silage(LBP group),50%hybrid Broussonetia papyrifera silage+50%corn silage(MBP group)and 100%hybrid Broussonetia papyrifera silage(BPS group)respectively.The pre-experiment lasted 15 days,and the experiment lasted 172 days.At the end of the experiment,six goats in each group were selected randomly for sampling.The rumen content was collected to measured rumen fermentation parameters and bacterial structure using the vacuum pump rumen fluid collector.The results showed that 6861 operational taxonomic units were obtained by 16S rRNA gene sequencing,which belonged to 230 genera of 20 phyla.The number of OTU,Chao1 index,Observed_species index,Shannon index of BPS group were 20.99%,27.23%,9.73%and 20.83%higher than those in the CS group,respectively(P<0.05).At the phylum level,Bacteroidetes,Firmicutes were the dominant flora in each group,the dominant flora of the genus were Prevotella,Bacteroidetes_unclassified and Ruminococcaceae_unclassified.The relative abundance of Bacteroidetes in BPS group was 15.89%and 20.17%lower than that in LBP group and MBP group respectively,the relative abundance of Prevotella in the BPS group was 39.86%and 39.25%lower than that in the CS group and the MBP group respectively(P<0.05).In summary,adding a high proportion of hybrid Broussonetia papyrifera silage can significantly improve the rumen flora richness of Wuchuan white goat.When 50%hybrid Broussonetia papyrifera silage was added,the relative abundance of Bacteroidetes and Prevotella was higher,which was beneficial to promoting the degradation of protein,starch and plant fiber in the rumen.
