摘要
目的观察柴术理胃饮对功能性消化不良大鼠胃肠运动及下丘脑组织中磷酸化p38丝裂原活化蛋白激酶(p-P38MAPK)、核转录因子(nuclear factor NF)-κB p65的影响,探讨其防治功能性消化不良的作用机理。方法 SPF级健康雄性SD大鼠60只,采用复合因素(慢性疲劳加悬尾加饮食失节)方法制备功能性消化不良模型,按随机数字表法随机分为空白组、模型组、柴术理胃饮低、中、高剂量组和多潘立酮组,每组10只。分别给予相应药物,观察动物的一般状态,计算胃排空率和小肠推进率,HE染色法观察胃组织病理学改变,WesternBlot和RT-PCR法检测大鼠下丘脑组织中p-P38MAPK、NF-κB p65蛋白和基因表达。结果柴术理胃饮各剂量组皆可升高模型组大鼠胃排空率和小肠推进率(P<0.05或P<0.01);模型组大鼠下丘脑组织中p-P38MAPK、NF-κB p65的蛋白和基因表达与正常组相比显著升高(P<0.01);不同剂量的柴术理胃饮均能不同程度降低大鼠下丘脑组织中p-P38MAPK、NF-κB p65蛋白和基因表达(P<0.01)。结论柴术理胃饮通过改善肝郁脾虚型功能性消化不良大鼠的胃排空功能、小肠推进功能和降低大鼠下丘脑组织中p-P38MAPK、NF-κB p65蛋白和基因表达来治疗功能性消化不良。
Objective To observe the effect of Chaizhu Liwei Yin on gastrointestinal motility and phosphorylation of p38 mitogen-activated protein kinase (p-P38MAPK) and nuclear factor-κB p65 (NF-κB p65) in the hypothalamus of rats with functional dyspepsia,and to explore its mechanism on the prevention and treatment of the disease.Methods A total of 60 SPF healthy male SD rats were used to establish a functional dyspepsia model by the method of compound factors (chronic fatigue,tail suspension and diet defect),and they were randomly divided into blank group,model group,Chaizhu Liwei Yin low-dose,medium-dose and high-dose groups and domperidone group,with 10 rats in each group.Corresponding drugs were respectively given to each group.The general state of the animals was observed,and the gastric emptying rate and small intestine propulsion rate were calculated,and the gastric pathological changes of stomach tissues were observed by HE staining,and the protein and gene expressions of p-P38MAPK and NF-κB p65 in the rat hypothalamus were detected by WesternBlot and RT-PCR.Results The gastric emptying rate and small intestine propulsion rate in the three-dose model groups were all significantly increased (P<0.05 or P<0.01).The protein and gene expressions of p-P38MAPK and NF-κB p65 in the hypothalamus of the model groups were significantly higher than that of the normal group (P<0.01).The protein and gene expression of p-P38MAPK and NF-κB p65 in the rat hypothalamus were decreased by different doses of Chaizhu Liwei Yin (P<0.01).Conclusion Chaizhu Liwei Yin can treat functional dyspepsia by improving the gastric emptying function and the small intestine propulsion function and decreasing the protein and gene expression of p-P38MAPK and NF-κB p65 in the hypothalamus of functional dyspepsia rats with the pattern of liver qi stagnation and spleen deficiency.
