摘要
目的:建立测定奥卡西平活性代谢产物10,11-二氢-10-羟基卡马西平血药浓度的超高效液相色谱法。方法:采集静脉血样本、高氯酸沉淀蛋白,离心取上清液进样,色谱柱为Waters ACQUITY UPLCBEH C_(18)(2.1 mm×50 mm, 1.7μm),流动相为0.05%甲酸∶乙腈=75∶25,检测波长254 nm,流速0.20 mL/min,柱温40℃。结果:10,11-二氢-10-羟基卡马西平在5~100μg/mL浓度范围峰面积与内标峰面积比值对浓度比值的线性关系良好(r=0.999 9),回收率、精密度、稳定性、准确度均在限度要求范围内。结论:该方法操作简单、准确、灵敏,可用于临床监测奥卡西平活性代谢产物10,11-二氢-10-羟基卡马西平血药浓度监测。
Objective:To establish the ultra-high performance liquid chromatography method for the blood concentration determination of active metabolite of oxcarbazepine 10,11-dihydro-10-hydroxyl carbamazepine.Methods:Venous blood samples were collected,proteins were precipitated by perchloric acid,and the supernatant was centrifuged and fed into the sample.The determination was performed on Waters ACQUITY UPLCBEH C_(18)(2.1 mm×50 mm,1.7μm)with 0.05%formic acid aqueous solution∶acetonitrile=75∶25 as mobile phase.The detection wavelength was 254 nm,the flow rate was 0.20 mL/min,and the column temperature was 40℃.Results:The linearity of the peak area of 10,11-dihydro-10-hydroxyl carbamazepine in the concentration range from 5 to 100μg/mL and the peak area ratio of the internal standard to the concentration ratio were good(r=0.999,9),and the recoveries,precision,stability and accuracy were within the limit requirements.Conclusion:The method is simple,accurate and sensitive,which can be used for blood concentration monitoring of active metabolite of oxcarbazepine 10,11-dihydro-10-hydroxyl carbamazepine.
