摘要
目的对比芪苓益肾颗粒传统制备工艺(TPP)与中药配方颗粒(CFG)化学成分种类和含量的差异。方法建立芪苓益肾颗粒指纹图谱,从相似度、化学成分种类、指标性成分含量、共有峰峰面积4个方面研究芪苓益肾颗粒TPP与CFG样品的差异。结果芪苓益肾颗粒TPP样品、A厂及B厂CFG样品相似度较高(0.959、0.956、0.926),C厂CFG样品相似度稍低(0.894)。TPP和CFG有20个(峰1~13,峰15~21)共有峰;TPP与CFG相比有峰14的出现及峰22的消失。测定了毛蕊异黄酮葡萄糖苷、阿魏酸、芒柄花苷、毛蕊异黄酮、芒柄花素5种指标成分的含量,发现CFG中各成分含量相对TPP均有不同程度降低。CFG共有峰总峰面积均低于TPP,A、B、C三厂CFG相对TPP共有峰总峰面积相对值分别为0.89、0.77、0.72。结论芪苓益肾颗粒TPP与CFG相比有化学成分的变化,TPP样品指标成分含量高于CFG样品;两者在化学成分种类、数目及指标成分含量均存在一定差异,表明CFG的标识当量存在一定误差,在临床应用中应进行修正,以促进临床合理应用。
Objective To compare the difference in the type and amount of chemical components between traditional preparation process(TPP)and Chinese medicine formula granules(CFG)of Qiling Yishen granules.Methods Fingerprints of Qiling Yishen granules were established to determine the difference in TPP and CFG from similarity,type of chemical,content of representative component,and common peak.Results The similarity of TPP samples,CFG samples A and CFG sample B was high(0.959,0.956,and 0.926),while of CFG samples C was slightly low(0.894).TPP and CFG had 20 common peaks(peak 1~13;peak 15~21).Compared with the CFG,peak 14 appeared and peak 22 disappeared in TPP.The content of 5 index components(verbasin glucoside,ferulic acid,formononetin,verbasin,and formononetin)was determined.The content of each component in CFG decreased compared with TPP.The total areas of CFG shared peaks were smaller than those of the TPP.Based on the total areas of TPP shared peaks,the of CFGs from factories A,B,and C were 0.89,0.77,and 0.72.Conclusion The components are changed in Qiling Yishen granules TPP and CFG sample.The content of index components in TPP sample is higher than that in CFG sample.There exist certain differences in the chemical component type,number and index component content.There is error in the labeling equivalent of CFG,which should be revised in clinical application.
