基于转录组学芯片数据和网络药理学预测龙葵干预胃癌的活性成分和作用机制 被引量:2

Prediction of Active Components and Mechanism of Solanum Nigrum Intervention in Gastric Cancer Based on Transcriptome Microarray Data and Network Pharmacology

摘要 目的:基于转录组芯片数据和网络药理学探讨龙葵治疗胃癌的有效成分和作用机制。方法:应用GEO下载胃癌相关转录组数据,TCMSP数据库检索龙葵活性成分和作用靶点,二者交集得到核心基因,生信网站绘制中药-成分-靶点桑基图,string网站绘制蛋白互作用图,DAVID对核心基因GO和KEGG富集分析。结果:GEO数据库检索数据集GSE118916,筛选到正常组和胃癌组织差异基因1056个;龙葵富含12个活性成分及靶基因137个,交集运算得到龙葵治疗胃癌的NR3C1等17个核心基因,活性成分与核心基因匹配桑基图显示了中药多成分多靶点的特点;GO和KEGG富集分析显示龙葵治疗胃癌涉及细胞对脂多糖的反应、肿瘤坏死因子信号通路等生物过程。结论:龙葵治疗胃癌的重要活性成分为槲皮素、谷固醇等,涉及TNF、IL16等信号通路。 Objective:To study the effective components and mechanism of Solanum nigrum in the treatment of gastric cancer based on transcriptional microarray data and network pharmacology.Methods:The data of gastric cancer related transcriptional group were downloaded by GEO,the active components and target points of Solanum nigrum were retrieved by TCMSP database.The core genes were collected by the intersection of the two.The Sangji map of traditional Chinese medicine component target point was drawn by Shengxin website,the protein interaction map was drawn on string website,and the enrichment and analysis of core genes go and KEGG by David were made by David.Results:The data set GSE118916 was retrieved from GEO database,1056 genes were screened from normal group and gastric cancer tissue;the results show that the active components and core genes match the mulberry base map of the active components and core genes;the enrichment analysis of GO and KEGG showed that the treatment of gastric cancer by anemone involves the cell response to lipopolysaccharide and tumor necrosis factor signaling pathway.Conclusion:Quercetin and glutaralol are the important active components in the treatment of gastric cancer,involving TNF,IL16 and other signaling pathways.
