黑菊芋多糖的提取工艺优化及其体外益生活性的研究 被引量:2

摘要 采用正交试验设计,结合氯仿-正丁醇法(Sevage法)、大孔树脂吸附及DEAE纤维柱法,优选黑菊芋多糖提取工艺;以红外光谱对多糖进行表征;并对黑菊芋多糖的组成进行分析;通过乳杆菌离体培养的方法研究黑菊芋多糖体外益生活性。结果表明:较优提取条件为超声温度80℃、超声时间60 min、料液比1∶15(g/mL)。黑菊芋多糖组成以果糖、葡萄糖、蔗糖和低聚果糖为主,含量分别为11.81、1.86、9.51、15.77 g/100 g。黑菊芋多糖分子中含有糖环结构和β-吡喃糖苷键。去除单糖的黑菊芋多糖对植物乳杆菌菌株ZL9和干酪乳杆菌菌株GL6的益生指数(prebiotic index,PI)值均在0.80以上。黑菊芋多糖可有效促进植物乳杆菌菌株ZL9和干酪乳杆菌菌株GL6的增殖,具有较好的体外益生活性。 This study aimed to optimize the purification process of polysaccharides from black jerusalem artichoke.An orthogonal experimental design was used to optimize the extraction process by combining the Sevage,macroporous resin,and DEAE fiber column methods.The polysaccharides in black jerusalem artichoke were characterized using infrared spectral analysis and the carbohydrate composition was determined.Probiotics activity of black jerusalem artichoke polysaccharides was studied by Lactobacillus in vitro culture.The optimum conditions of extraction were 80°C,60 min,and 1∶15(g/mL).Infrared spectral analysis indicated the presence of a sugar ring structure andβ-glucopyranoside bonds in the black jerusalem artichoke polysaccharides.Fructose,glucose,sucrose,and fructooligosaccharide were the main components with concentrations of 11.81,1.86,9.51 g/100 g,and 15.77 g/100 g.The prebiotic index of black jerusalem artichoke polysaccharide without monosaccharide on Lactobacillus plantarum strain ZL9 and Lactobacillus casei strain GL6 was above 0.80,which can effectively promote the proliferation of both strains and showed good prebiotic activity in vitro.
