针康法对脑缺血大鼠缺血区BDNF、GFAP和Caspase-3基因表达的影响 被引量:2

Effect of the acupuncture and rehabilitation therapy on the gene expression of BDNF,GFAP and Caspase-3 in the ischemic area of rats with cerebral ischemia

摘要 目的探讨针康法对脑缺血大鼠缺血区BDNF、GFAP和Caspase-3基因表达的影响。方法雌性SD大鼠50只随机分为空白组、模型组、针灸组、康复组和针康组,每组各10只。空白组大鼠给予假手术处理,模型组大鼠采用Longa改良线栓法复制脑缺血模型;针灸组在模型组基础上给予头穴从刺法,每日1次;康复组在模型组基础上给予跑台训练,每日1次;针康组在模型组基础上给予每日1次头穴从刺法和跑台训练。所有大鼠术后治疗14 d,收集脑组织样品,qRT-PCR法检测BDNF、GFAP和Caspase-3 mRNA表达情况,Western blot检测BDNF、GFAP和Caspase-3蛋白表达情况。结果与空白组相比,模型组BDNF、GFAP和Caspase-3基因表达显著增加(P < 0.05);与模型组比较,针灸组、康复组和针康组BDNF和GFAP 基因表达水平显著增加(P<0.05),而Caspase-3基因表达显著减少(P<0.05);与针灸组和康复组比较,针康组BDNF和GFAP 基因表达水平显著增加(P< 0.05),而Caspase-3基因表达显著减少(P<0.05)。结论单纯针灸或单纯康复疗法均可有效抑制缺血区细胞凋亡、保护神经元并促进其功能修复;与此相比,针康疗法能更好地抑制细胞凋亡,保护缺血区受损脑细胞,更利于脑缺血区功能重建。 Objective To investigate the effect of the acupuncture and rehabilitation therapy on the gene expression of BDNF,GFAP and Caspase-3 in the ischemic area of rats with cerebral ischemia.Methods A total of fifty female SD rats were randomly divided into a blank group,a model group,an acupuncture group,a rehabilitation group and an acupuncture-rehabilitation group,with 10 rats in each group.The rats in the blank group were treated with a sham operation,while the rats in the model group were treated with the modified Longa suture method to replicate the cerebral ischemia model,and the acupuncture group was given scalp acupuncture on the basis of the model group once a day,while the rehabilitation group was given the treadmill training on the basis of the model group once a day,and the acupuncture-rehabilitation group was given the daily scalp acupuncture and treadmill training on the basis of the model group.After 14 days of postoperative treatment,the gene expression levels of BDNF,GFAP and Caspase-3 mRNA were detected by qRT-PCR,while the protein expression levels of BDNF,GFAP and Caspase-3 were detected by Western blot.Results Compared with the blank group,the gene expression levels of BDNF,GFAP and Caspase-3 in the model group were increased significantly (P<0.05).Compared with the model group,the gene expression levels of BDNF and GFAP in the acupuncture group,rehabilitation group and acupuncture-rehabilitation group were increased significantly (P<0.05),while the gene expression level of Caspase-3 was significantly decreased (P<0.05).Compared with the acupuncture group and rehabilitation group,the gene expression levels of BDNF and GFAP in the acupuncture-rehabilitation group were significantly increased (P<0.05),while the gene expression level of Caspase-3 was significant decreased (P<0.05).Conclusion Acupuncture alone or the rehabilitation therapy alone can effectively inhibit apoptosis,protect neurons and promote their functional repair in the ischemic area.While compared with that,the acupuncture-rehabilitation therapy can better inhibit apoptosis and protect damaged brain cells in the ischemic area and be more conducive to the functional reconstruction of cerebral ischemic areas.
