摘要
目的:探讨细菌脂多糖(Lipopolysaccharide,LPS)对细菌性脓毒症树突状细胞(Dendritic Cells,DCs)的胞饮作用及抗原交叉提呈作用的影响。方法:以脂多糖刺激骨髓来源树突状细胞,建立脓毒症免疫麻痹模型,采用流式细胞分析法检测树突状细胞对荧光黄(Lucifer Yellow,LY)的胞饮能力、共刺激分子CD80和CD86的表达水平以及抗原交叉提呈作用。结果:与对照组相比,模型组DCs胞饮LY阳性细胞百分率显著升高(P<0.01),CD80、CD86阳性细胞百分率显著升高(P<0.01),并且交叉提呈SIINFEKL-H2Kb的阳性百分率显著升高(P<0.001)。结论:脂多糖可上调树突状细胞免疫共刺激分子CD80、CD86表达,显著促进树突状细胞的胞饮作用以及抗原交叉提呈能力。
Objective:To investigate efficacy of LPS on the pinocytosis and antigen cross-presentation of DCs in bacterial sepsis.Methods:A sepsis immune paralysis model was established by LPS after stimulating bone marrow-derived DCs.The LY pinocytosis level,the expression of costimulatory molecules CD80 and CD86 and antigen-specific cross presentation of DCs induced by lipopolysaccharide were detected by flow cytometry.Results:Compared with the control group,the percentage of LY positive cells and the percentage of CD80 and CD86 positive cells in the model group were both significantly increased.The positive rate of cross presentation of SIINFEKLH2Kb in the model group was significantly increased.Conclusion:LPS up-regulates the expression of immunostimulatory molecules CD80 and CD86,and significantly promotes pinocytosis and antigen cross-presentation of DCs.
