摘要
N6-甲基腺苷(N6-methyladenosine,m^(6)A)修饰是指mRNA中腺苷酸(A)的第六位氮原子处发生甲基化,m^(6)A修饰是生物mRNA中最广泛存在的内部修饰。m^(6)A修饰通过调节RNA代谢、稳定性、翻译、降解以及选择性剪接来发挥生物学功能,在细胞分化、生物节律、生殖生理、肿瘤发生等方面起到调控作用。本文综述了目前对m^(6)A修饰的认识,阐述了甲基化转移酶、去甲基化酶、甲基化阅读蛋白3种m^(6)A甲基化酶的作用,总结了m^(6)A修饰在调节动物生殖方面的研究进展,为m^(6)A甲基化在家畜生殖生理方面的相关研究与应用提供参考。
N6-methyladenosine(m^(6)A)refers to a methylation at the sixth nitrogen atom of adenosine(A)in messenger RNAs(mRNAs).The m^(6)A modification is the most prevalent internal modification in mRNA of all higher eukaryotes.The m^(6)A modification plays a biological role by regulating RNA metabolism,stability,translation,degradation and selective splicing,and plays a regulatory role in cell differentiation,biological rhythm,reproductive physiology and tumorigenesis.In this paper,we reviewed the current understanding of m^(6)A methylation,and described the functions of writer,eraser and reader.Besides,the research progress on m^(6)A methylation in regulating animal reproduction were also be summarized.The aim of this review is to provide technical material and theoretical reference for the research and application regarding with the m^(6)A methylation in reproductive physiology of domestic animals.
