山东省养鸡场大肠杆菌多黏菌素耐药基因mcr-1的流行情况 被引量:3

Prevalence of polymyxin-resistant gene mcr-1 in Escherichia coli from chicken farms in Shandong Province

摘要 2017-2018年,从山东省8个大型集约化养鸡场中共采集720份新鲜粪便样品,通过细菌分离鉴定共分离到697株非重复大肠杆菌。采用琼脂稀释法对所有菌株进行药敏试验;利用PCR扩增多黏菌素耐药基因(mcr-1);并对mcr-1阳性菌株扩增β-内酰胺类耐药基因(ESBL)和喹诺酮类耐药基因(PMQR)。结果显示,从697株大肠杆菌中共检测到83株(11.9%)携带mcr-1基因,其中mcr-1阳性菌株对氨苄西林的耐药率最高(82/83,98.8%),其次是阿莫西林(81/83,97.6%)和氟苯尼考(79/83,95.2%),但对阿莫西林/克拉维酸的耐药率较低(18/83,21.7%)。从mcr-1阳性菌株中共检测到5种ESBL基因,其中所有菌株均携带bla;(83/83,100%),其次为bla;(67/83,80.7%);检测到4种PMQR基因,其中aac(6′)-Ib-cr检出率最高(34/83,40.9%),其次为qnrA(9/83,10.8%)。结果表明,山东省禽源大肠杆菌是mcr-1基因的储存库,可能对人类公共健康造成威胁,应继续加强对养鸡场中mcr-1耐药基因的监控。 This study aims to understand the prevalence of polymyxin-resistant gene mcr-1 in Escherichia coli(E.coli)from chicken farms in Shandong Province.From 2017 to 2018,a total of 720 fresh fecal samples were collected from 8 large-scale intensive chicken farms in Shandong Province,and 697 strains of non-repetitive E.coli were isolated through bacterial isolation and identification.Drug sensitivity test for all E.coli was performed using micro-broth dilution method.PCR was used to amplify the polymyxin resistant gene mcr-1 and mcr-1 positive isolates were PCR-screened forβ-lactam resistance gene(ESBL)and quinolone resistance gene(PMQR).The results showed that 83(11.9%)of 697 E.coli harboured the mcr-1 gene.The resistance rate of mcr-1 positive strains to ampicillin was the highest(82/83,98.8%),followed by amoxicillin(81/83,97.6%)and florfenicol(79/83,95.2%),however,the resistance rate to amoxicillin/clavulanic acid was low(18/83,21.7%).Five ESBL genes were detected from mcr-1 positive strains,of which bla;(83/83,100%)was carried by all strains,followed by bla;(67/83,80.7%);four PMQR genes were detected,among which aac(6′)-Ib-cr was the highest(34/83,40.9%),followed by qnrA(19/83,10.8%).In conclusion,E.coli of chicken in Shandong Province is the repository of mcr-1 gene,which may pose a threat to human public health.We should continue to strengthen the monitoring of mcr-1 resistance gene in chicken farms.
