响应面法优化羊源解淀粉芽孢杆菌冻干保护剂配方 被引量:5

Optimization of cryoprotectant formulation for Bacillus amyloliquefaciens isolated from goat by response surface methodology

摘要 为制备羊源解淀粉芽孢杆菌fsznc-06菌.剂,采用响应面法,分析优化冻干保护剂配方,从离心条件、保护剂筛选与复配、稳定性检测等方面探究对菌体存活率的影响.结果表明,最佳离心条件为6000 r/min离心10 min,优化后的冻干保护剂配方为丙三醇(3.102 g/100 mL)、蔗糖(2.926 g/100 mL)和葡萄糖(7.096 g/100 mL),在该条件下解淀粉芽孢杆菌存活率达到96.47%,与理论预测值接近.冻干菌剂在4℃条件下贮存稳定性更高,保藏180天后的存活率约为66.30%,说明响应面法可用于优化解淀粉芽孢杆菌冻干保护剂. To prepare Bacillus amyloliquefaciens fsznc-06 from goat feces,cryoprotectants were optimized by response surface analysis.The effects on the survival rate of bacteria were explored from the aspects of centrifugal conditions,selection,formulation of cryoprotectants and stability detection.The results showed that the optimal centrifugation conditions were 6000 r/min for 10 min.The optimized cryoprotectants contained glycerol(3.102 g/100 mL),sucrose(2.926 g/100 mL),and glucose(7.096 g/100 mL),and its lyophilization survival rate reached 96.47%,which was close to the theoretical prediction.The freeze-dried bacterial agent had higher storage stability at 4℃,and the survival rate after storage for 180 days was about 66.30%.Therefore the response surface method can be used to optimize the cryoprotectant of Bacillus amyloliquefaciens.
