摘要 目的:采用Box-Behnken响应面法优化满山红油脂质体处方。方法:采用乙醇注入-探头超声制备满山红油脂质体,HPLC法测定吉马酮的含量,以卵磷脂与胆固醇的比例(X_(1))、脂药比(X_(2))、磷脂浓度(X_(3))作为自变量,包封率(Y)作为因变量,根据Box-Behnken试验的设计原理,以包封率为指标对各个因素进行优化,并对包封率进行预测分析,对制备的满山红油脂质体包封率、粒径、多分散指数(PDI)、Zeta电位和载药量进行质量评价。结果:满山红油脂质体最佳处方为X_(1)=7.28∶1、X_(2)=11.34∶1、X_(3)=9.32 mg/mL,包封率(Y)为(82.55±1.66)%,粒径为(130.53±24.46)nm,多分散指数为0.185±0.050,Zeta电位为(21.97±1.36)mV,载药量为(5.94±0.12)%。结论:采用Box-Behnken响应面法能够快速、准确地得到满山红油脂质体的优化处方,预测效果较好,满山红油脂质体制备工艺稳定可行。 Objective:To optimize the formulation of Rhododendri Daurici oil liposomes using Box-Behnken response sur-face methodology.Methods:Ethanol injection-probe ultrasound was used to prepare Rhododendri Daurici oil liposomes,and the content of gemmazone was determined by HPLC.The ratio of lecithin to cholesterol(X_(1)),lipid-drug ratio(X_(2)),and phospholipid concentration(X_(3))were used as independent variables,and the encapsulation rate(Y)was used as the dependent variable.Ac-cording to the design principle of the Box-Behnken test,the encapsulation rate was used as the index to optimize each factor for the index,to predict and analyze the entrapment efficiency,and to evaluate the entrapment efficiency,particle size,polydispersity in-dex(PDI),Zeta potential and drug loading of the prepared Rhododendri Daurici oil liposomes.Results:The best prescription of Rhododendri Daurici oil liposome was X_(1)=7.28∶1,X_(2)=11.34∶1,X_(3)=9.32 mg/mL,the encapsulation efficiency(Y)was(82.55±1.66)%,and the particle size was(130.53±24.46)nm,the polydispersity index was 0.185±0.050,the zeta potential is(21.97±1.36)mV,and the drug loading was(5.94±0.12)%.Conclusion:The Box-Behnken response surface method can quickly and accurately obtain the optimized prescription of the Rhododendri Daurici oil liposomes,and the prediction effect is good.The preparation process of the Rhododendri Daurici oil liposomes is stable and feasible.
