摘要
目的探讨益景汤对糖尿病(DM)大鼠血-视网膜屏障(BRB)的保护效应及机制。方法45只大鼠被随机分为3组:正常组、糖尿病组(DM组)、益景汤组(YJ组),每组15只,采用链脲佐菌素对DM组、YJ组大鼠进行DM造模。正常组和DM组采用PBS进行灌胃干预,YJ组采用益景汤灌胃干预。16周后处死所有大鼠,采用伊凡思蓝检测BRB渗漏,Western blot法检测视网膜组织occludin、claudin-5、ZO-1等紧密连接相关蛋白的表达。结果(1)视网膜血管通透性:在第16周末时,DM组大鼠视网膜微血管管径欠均匀、管迹模糊,存在明显的微血管渗漏,而YJ组大部分视网膜微血管管径均匀、管迹清晰,少量视网膜微血管渗漏。(2)紧密连接相关蛋白的表达:DM组大鼠视网膜中occludin、claudin-5、ZO-1等紧密连接相关蛋白表达量降低,与正常组相比差异均有统计学意义(t_(occludin)=23.568,t_(claudin)-5=17.447,t_(ZO-1)=21.365,均P=0.000)。益景汤能抑制DM模型大鼠视网膜中occludin、_(claudin)-5、_(ZO-1)蛋白的表达量下降,YJ组上述蛋白的表达量与DM组相比差异具有统计学意义(t_(occludin)=5.539,t_(claudin)-5=5.466,均P=0.000,t_(ZO-1)=3.325,P=0.008)。结论益景汤能够抑制DM大鼠视网膜组织中occludin、claudin-5、ZO-1等紧密连接相关蛋白表达量下降,降低BRB的损害,减轻DM大鼠视网膜微血管的渗漏,对早期DR具有防治作用。
OBJECTIVE To investigate the protective effect and mechanism of Yijing Decoction on blood-retinal barrier(BRB)in rats with diabetes mellitus(DM).METHODS Fourty-five rats were randomly divided into 3 groups:normal group(NG),DM group(DMG)and Yijing Decoction group(YJG)(15 rats in each group).Streptozotocin was used to make DM models in DMG and YJG.The NG and DMG were treated with PBS,and YJG was administered by Yijing Decoction.After 16 weeks,all rats were sacrificed to assess the leakage of BRB by Evans blue,and the expression of tight junction related proteins such as occluding,claudin-5 and ZO-1 in retinal tissues was detected by Western blot.RESULTS(1)Retinal vascular permeability:At the end of 16th week,retinal microvessels of DMG demonstrated uneven diameters,obscure vascular tracts,and obvious retinal microvascular leakage,while most retinal microvessels of YJG had uniform diameters,clear traces,and a small amount of retinal microvascular leakage.(2)Expression of tight junction related proteins:The protein expression of occludin,claudin-5,ZO-1 in the retina of DMG was significantly lower than that of NG,and the difference was statistically significant(t_(occludin)=23.568,t_(claudin)-5=17.447,t_(ZO-1)=21.365,all P=0.000)Yijing Decoction can inhibit the decrease of the expression of these proteins in the retina of DM rats,and the difference was statistically significant compared with DMG(t_(occludin)=5.539,P=0.000;t_(claudin)-5=5.466,all P=0.000;t_(ZO-1)=3.325,P=0.008).CONCLUSIONS Yijing Decoction can inhibit the decrease of the protein expression of occludin,claudin-5,ZO-1 in the retina of DM rats,alleviate BRB injury,and reduce the leakage of retinal microvessels in DM rats,so as to prevent and treat DR in early stage.
