创伤后应激障碍模型大鼠的强迫性和攻击性行为观察

Observation on the obsessive-compulsive aggression behaviors in model rats with post-traumatic stress disorder

摘要 目的:探究创伤后应激障碍(PTSD)模型大鼠的强迫性和攻击性行为并分析其关联性,为精神病司法鉴定程序启动标准的调整提供可能的实验参考。方法:选取24只SD大鼠(体质量180~220 g)随机分为正常组(n=8)、PTSD组(n=9)、强迫症(OCD)组(n=7);另取20只21天龄SD大鼠随机均分为攻击组、工具鼠组。PTSD组、OCD组、攻击组大鼠分别用单次延长应激、喹吡罗、应激源性攻击方法造模,通过旷场和高架十字迷宫、离巢返巢、居住入侵与拒缚反射等试验进行相应模型鉴定和行为观察。结果:①与正常组相比,PTSD组在旷场试验中总路程、跨格数、中央区域停留时间均减少,在高架十字迷宫试验中总路程、进入开放臂时间均减少,在离巢返巢试验中运动总路程减少,回家停留时间增加。②在居住入侵试验与拒缚反射评分中,PTSD组、OCD组的评分相近,均高于正常组低于攻击组。③PTSD组、OCD组的强迫性与攻击性行为之间均存在正相关。结论:PTSD大鼠具有明显的强迫性行为与攻击性行为,且两者间可能存在关联性。 Objective:To investigate the obsessive-compulsive aggressive behavior in model rats with post-traumatic stress disorder(PTSD)and the correlation of PTSD with the aggressive behavior for potential empirical reference to modify the criteria in initiation of justicial appraisal.Methods:Twenty-four SD rats,weighed 180-220g,were randomized to normal group(n=8),PTSD group(n=9)and obsessive-compulsive disorder(OCD)group(n=7).Another 20 SD rats(21 days old)were evenly divided into aggressive behavior(PA)group and control rat group.The PTSD rats were prepared by single-prolonged stress and evaluated by the open-field test and elevated plus maze test.The OCD rats were injected with quinpirole and identified by obsessive checking home test.The PA rats were prepared by stress-derived attack method and evaluated by the resident-intruder test and the bound refusing reflection score.Results:①Compared with the normal rats,the total movement distance,zone transition number and resting time in the central area of the PTSD rats were decreased in the open field test.The total movement distance and the percentage of time into the open arm of the PTSD rats were also decreased in the elevated cross maze test;②In the obsessive checking home test,the PTSD rats moved less distance in total,and stayed longer in home than the normal rats;In the resident-intruder test and the bound refusing reflection score,the PTSD and OCD rats were similar,which were both higher than the normal rats yet lower than the aggression rats;③Obsessive-compulsive and aggressive behaviors were in positive correlation in rats of PTSD and OCD groups.Conclusion:Obsessive-compulsive and aggressive behaviors are dominant in PTSD rats,suggesting that there may be a relationship between these behaviors.
