摘要
采用硅胶柱色谱、 Sephadex LH-20凝胶柱层析、 C18反向色谱柱和制备液相色谱等方法对云南产白花紫苏(Coleus esquirolii)的乙醇提取物进行了分离纯化,并通过核磁共振、质谱等现代波谱学技术对所分离得到的化合物进行了结构鉴定。从云南产白花紫苏中分离和鉴定了11个化合物:毛萼鞘蕊花乙素(1), cariocal (2),白桦脂醇(3),白桦脂酸(4),麦角甾-6, 22双烯-5α,8α-表二氧-3β-醇(5),芫花素(6),芹菜素(7),山奈酚(8),槲皮苷(9),黄芪苷(10)和杨梅树皮苷(11)。并对分离的黄酮化合物进行了抗氧化活性研究,研究表明白花紫苏中主要以萜类和黄酮类化合物为主,其中化合物3、 4、 5、 6、 7、 8、 9、 10和11为首次从白花紫苏中分离得到;化合物9和11清除DPPH自由基的半数抑制率分别为(19.51±0.03)μM和(23.13±0.05)μM,显现了较强的抗氧化活性。
The chemical constituents ofColeus esquiroliiwere isolated by silica gel, Sephadex LH-20 and C18 reversed chromatography, and their structures were assigned by nuclear magnetic resonance, mass spectrometry, and infrared spectroscopy etc. Eleven compounds were isolated, and their structures were identified as esquirolin B(1), cariocal(2), botulin(3), betulinic acid(4),5α, 8α-epidioxy-6, 22-diene-3β-ol(5), genkwanin(6), apigenin(7), kaempferol(8), quercitrin(9), astragalin(10) and myricitrin(11). Furthermore, antioxidant of flavonoids which isolated from the plant were evaluated. Results showed that terpenoids and flavonoids were the main compounds in the Coleus esquirolii, and compound 3, 4, 5, 6, 7, 8, 9, 10 and 11 were obtained from this plant for the first time. The IC50 of compounds 9 and 11 in scavenging DPPH free radicals were(19. 51 ± 0. 03) μM and(23. 13 ± 0. 05) μM, respectively, which exhibit significant antioxidant activities.
