B7-H4、miR-145和β-连环素在结直肠癌组织中的表达及相关性研究 被引量:2

摘要 目的研究T细胞共刺激分子B7(T cells co-stimulate the molecule B7,B7-H4)、microRNA-145(miR-145)、β-连环素在结直肠癌组织中的表达及相关性。方法选取2019年1月至2020年12月手术后结直肠癌组织标本和癌旁组织标本各48例,对2组组织中B7-H4、miR-145、β-连环素表达进行检测,并按照WHO(1999)大肠肿瘤组织学分型标准进行划分,分析B7-H4、miR-145、β-连环素与不同分化之间的关系;根据国际抗癌联盟(UICC)1997年修订的大肠癌Dukes分期标准进行划分,分析B7-H4、miR-145、β-连环素与不同分期之间的关系,采用Pearson分析B7-H4、miR-145、β-连环素之间的相关性。采用免疫组化染色对B7-H4、miR-145、β-连环素表达进行检测。结果与癌旁正常组织比较,结直肠癌组B7-H4、β-连环素表达显著升高,miR-145表达显著降低,差异有统计学意义(P<0.05);与无淋巴结转移比较,有淋巴结转移B7-H4、β-连环素表达显著升高,miR-145表达显著降低,差异有统计学意义(P<0.05);与高分化比较,中分化、低分化B7-H4、β-连环素表达显著升高,miR-145表达显著降低(P<0.05);与中分化比较,低分化B7-H4、β-连环素表达显著升高,miR-145表达显著降低(P<0.05);与Ⅰ期比较,Ⅱ期、Ⅲ~Ⅳ期B7-H4、β-连环素表达显著升高,miR-145表达显著降低,差异有统计学意义(P<0.05);与Ⅱ期比较,Ⅲ~Ⅳ期B7-H4、β-连环素表达显著升高,miR-145表达显著降低,差异有统计学意义(P<0.05)。B7-H4、miR-145、β-连环素之间进行相关性分析,B7-H4、miR-145之间均呈负相关(r=-0.481,P=0.001);miR-145、β-连环素之间均呈负相关(r=-0.672,P=0.001);B7-H4、β-连环素之间均呈正相关(r=0.609,P=0.001)。结论B7-H4、β-连环素在结肠癌组织中高表达,miR-145低表达,与癌细胞的分化程度、分期密切相关,三项联合检测可成为检测结直肠癌的有效指标。 Objective To investigate the expression and correlation of T cells co-stimulating molecule B7(B7-H4),microRNA-145(miR-145)and beta-catenin in colorectal cancer tissues.Methods A total of 48 cases of colorectal cancer tissues and adjacent tissues by surgical excision from January 2019 to December 2020 were collected,and the expression levels of B7-H4,Mir-145 and beta-catenin were detected.The correlation between B7-H4,Mir-145,beta-catenin and different differentiation as well as different stages was analyzed according to WHO(1999)and Dukes staging criteria for colorectal cancer revised by the International Union against Cancer(UICC)in 1997.Pearson analysis was used to analyze the correlation among B7-H4,miR-145,catenin.Immunohistochemical staining was used to detect the expression levels of B7-H4,miR-145 and-catenin.Results Compared with those in adjacent normal tissues,the expression levels of B7-H4 and beta-catenin in colorectal cancer tissues were significantly increased,however,the expression levels of Mir-145 were significantly decreased(P<0.05).Compared with those in no lymph node metastasis,the expression levels of B7-H4 and beta-catenin in lymph node metastasis were significantly increased,but those of miR-145 were significantly decreased(P<0.05).Compared with thosein high differentiation tissues,the expression levels of B7-H4 and beta-catenin in moderately and poorly differentiated tissues were significantly increased,while those of miR-145 were significantly decreased(P<0.05).Compared with those at stageⅡ,the expression levels of B7-H4,beta-catenin at stageⅢ~Ⅳwere significant increased,but those of miR-145 were significantly decreased(P<0.05).There was a negative correlation Between B7-H4 and Mir-145 as well as between Mir-145 and beta-catenin(P<0.01),however,there was a positive correlation between B7-H4 and-catenin(P<0.01)Conclusion High expression of B7-H4 and beta-catenin in colon cancer tissues and low expression of Mir-145 are closely related to the differentiation degree and stage of cancer cells.The combined detection of the three indexes can be used in diagnosis of colorectal cancer.
