摘要
试验选用副干酪乳杆菌SLP16研发新型限抗饲料添加剂,以麸皮为固体发酵基质,通过单因素和正交试验对副干酪乳杆菌SLP16发酵条件进行优化。结果显示,在接种量16%、液料比1.2 mL/g、37℃、发酵3 d的条件下优化后的发酵饲料添加剂能够有效抑制革兰氏阳性菌和阴性菌,对金黄色葡萄球菌的抑菌圈直径为25.0 mm,效价6.58 lg(IU/g);对大肠杆菌的抑菌圈直径为22.0 mm,效价为5.70 lg(IU/g)。60 d常温保存,发酵限抗饲料添加剂的抑菌能力超过95%,抑菌稳定性优异。研究表明,饲料添加剂经微生物发酵后,DPPH自由基清除率和还原力均明显增加。
The study used the Lactobacillus paracasei SLP16 to develop a novel anti-resistant feed additive, using wheat bran as a solid fermentation substrate. The fermentation conditions of Lactobacillus paracasei SLP16 were optimized by single factor and orthogonal experiments. The results showed that the optimized fermentation feed additive could effectively inhibit Gram-positive bacteria and Gram-negative bacteria at the inoculation rate of 16%, liquid-solid ratio of1.2 mL/g, fermentation at 37 ℃ for 3 d. Staphylococcus aureus was used as indicator bacteria, the inhibition zone diameter was 25.0 mm, and the titer was 6.58 lg(IU/g). Escherichia coli was used as the indicator bacteria, the inhibition zone diameter was 22.0 mm, and the titer was 5.70 lg(IU/g). When being stored at room temperature for 60 d, the bacteriostatic capacity of the feed additives was more than 95%, and the bacteriostatic stability was excellent. The experiment indicates that the scavenging rate of DPPH free radical and reducing power of feed additive are significantly increased after microbial fermentation.
