希瓦氏菌对甲基绿的脱色及其机制 被引量:2

Decolorization of methyl green by Shewanella oneidensis MR-1 and its mechanism

摘要 以希瓦氏菌(Shewanella oneidensis MR-1)为染料脱色菌株对甲基绿(methyl green,MG)进行脱色处理,研究不同碳源、甲基绿浓度、初始菌株浓度和温度对染料脱色的影响。通过紫外-可见吸收光谱(UV-visible)、傅里叶红外光谱(FTIR)以及液相色谱-质谱联用(LC-MS)对甲基绿的脱色产物进行分析研究,通过杀菌实验去测定甲基绿脱色液的生物毒性,通过S.oneidensis MR-1及其突变菌株对甲基绿的脱色速率推测可能的生物机制。研究结果表明:S.oneidensis MR-1在以甲酸钠为碳源时的脱色率最高;在不同甲基绿浓度下,该菌对40 mg·L^(-1)甲基绿可在1 h内脱色率达95%以上;菌株浓度的增加会增强脱色效果;温度研究显示,S.oneidensis MR-1在30℃条件时,对甲基绿的脱色效果更好。产物分析及杀菌实验结果证实S.oneidensis MR-1能够对甲基绿进行脱色,脱色后液体无毒,脱色产物主要为:4-(N,N-二甲基氨基)-4′-(N′,N′-二甲基氨基)二苯甲酮(DDBP),4-(N-乙基-N,N-二甲基氨基)-4′-(N′-甲基氨基)二苯甲酮(ED-MBP),4-(N-乙基-N,N-甲基氨基)-4′-(N′,N′-二甲基氨基)二苯甲酮(EM-DBP)和4-(N-乙基-N,N-甲基氨基)-4′-(N′-甲基氨基)二苯甲酮(ED-BP)。 The present study was conducted using Shewanella oneidensis MR-1 for the decolorization of triphenylmethane dye methyl green to evaluate the effects of various parameters(different carbon sources,initial concentrations of methyl green and S.oneidensis MR-1 and temperature)on methyl green removal.Decolorizing products of methyl green were analyzed by UV-visible,FTIR analysis and LC-MS analyses.The toxicity of methyl green decolorizing solution was determined by the killing assay.The possible biological mechanism was inferred from the decolorization rate of methyl green by S.oneidensis MR-1 wild-type and mutant strains.The results showed that S.oneidensis MR-1 had the highest decolorization rate when the carbon source was formic acid.Under different concentrations of methyl green,the decolorization efficiency was up to 95%after 1 h when methyl green concentration was 40 mg·L^(-1).The decolorization efficiency was enhanced with the increase of strain concentration.Temperature experiment showed that the decolorization efficiency of methyl green was better at 30℃.The results of product analysis and killing assay confirmed that S.oneidensis MR-1 could decolorize methyl green and the decolorizing solution had no obvious toxicity.The decolorizing products of methyl green included 4-(N,N-Dimethylamino)-4’-(N’,N’-dimethylamino)benzophenone(DDBP),4-(N-Ethyl-N,N-dimethylamino)-4’-(N’-methylamino)benzophenone(ED-MBP),4-(NEthyl-N-methylamino)-4’-(N’,N’-dimethylamino)benzophenone(EM-DBP)and 4-(N-Ethyl-N-methylamino)-4’-(N’-methylamino)benzophenone(ED-BP).
