薏苡仁麸炒前后治疗脾虚水湿不化谱效关系研究 被引量:9

Spectrum-effect relationship of Coicis Semen due to bran stir-frying in improving Spleen Deficiency and Water Dampness

摘要 目的研究薏苡仁麸炒前后HPLC指纹图谱与治疗脾虚水湿不化药效之间的相关性,为明确薏苡仁麸炒前后药效物质基础提供实验依据。方法通过HPLC法建立麸炒前后薏苡仁指纹图谱;建立脾虚水湿不化大鼠模型,比较其麸炒前后的药效作用;同时采用灰色关联度法,对5批生品及其麸炒品进行谱效关系分析,分别找出关联度较大的化学成分,并比较麸炒前后谱-效关系差异。结果 HPLC指纹图谱中指认9个共有峰。与模型组比较,生品与麸炒品均有显著性差异;与生品组比较,麸炒薏苡仁各药效指标明显高于生品。麸炒品中9种共有成分与GAS、MTL、SS、体质量、胸腺指数以及脾指数的关联度明显高于生品;生品中9种共有成分与VIP的关联度明显高于麸炒品。结论薏苡仁治疗脾虚水湿不化的药效是多成分共同作用结果,麸炒后药效明显高于麸炒前。 AIM To study the correlation between the HPLC fingerprints of Coicis Semen and the efficacy in the improvement of Spleen Deficiency and Water Dampness,and to clarify the material basis of the drug effect of Coicis Semen before and after bran stir-frying.METHODS Both HPLC fingerprints of Coicis Semen before and after bran stir-frying,and a rat model of Spleen Deficiency and Dampness were established.Five batches of raw Coicis Semen and their bran-fried products were subjected to the analyses by the grey correlation method for determination of the key chemical components in the spectrum-effect relationship and the efficacy of bran stir-frying.RESULTS Nine common peaks were identified in the HPLC fingerprints.Groups intervened with raw Coicis Semen and bran-fried Coicis Semen,with the latter demonstrating significantly better efficacy than the former as revealed by the levels of the assessment indices,were observed with significant difference in contrast to the model group.Nine common components in bran-fried Coicis Semen brought forth significantly higher levels of GAS,MTL,SS,body mass,thymus index and spleen index than those in the raw products who contributed significantly higher degree of VIP correlation.CONCLUSION With resultant improvement in Spleen Deficiency and Water Dampness because of multi-component interaction,Coicis Semen may find an even significantly better efficacy after bran stir-frying.
