摘要
景观水体作为城市水系的重要组成部分,氮素超标是影响其生态功能的主要因素.为了探究城市景观水体反硝化菌群落特征和驱动因素,利用nirS功能基因研究石家庄市主城区4个公园的景观水体和沉积物的反硝化菌群落分布特征,结合水体和沉积物氮素指标对景观水体反硝化菌群落演变的驱动因子进行分析.结果表明,景观水体水质参数季节差异显著(P<0.001),沉积物氮素指标空间差异显著(P<0.001);水体以及沉积物的α多样性季节和空间差异都不显著(P>0.05),电导率(EC)和强氧化剂浸取态氨氮(SOEF-NH+4-N)是影响水体以及沉积物的α多样性的主要环境因素;景观水体反硝化菌主要为变形菌门(Protebacterice)中的Dechloromonas、Rhodocyclaceae、Pseudomonas、Rhodobacter和Thauera;主坐标分析(PCoA)表明水体和沉积物反硝化菌群组成空间差异显著(P<0.001),水体关键反硝化物种也呈现出显著的空间差异(P<0.001);冗余分析(RDA)和随机森林分析(RF)指出,高锰酸盐指数和总磷(TP)是影响水体反硝化菌群和关键物种组成的主要环境因子,SOEF-NH+4-N、离子交换态氨氮(IEF-NH+4-N)和弱酸浸取态硝氮(WAEF-NO-3-N)是影响沉积物反硝化菌群和关键物种组成的主要环境因子.综上可知,通过对景观水体和沉积物反硝化菌群落分布特征和驱动因素的分析,为进一步认识景观水体氮素和反硝化菌群的互作机制提供借鉴.
Landscape water is an important part of urban water systems,and excessive nitrogen affects its ecological functions.This study aimed to investigate the temporal and spatial distribution characteristics and driving factors of the community structure of denitrifying bacteria from landscape water.The functional gene nirS was used as a functional marker to explore the community of denitrifying bacteria in the water and sediment of landscape water.Based on parameters of the water and sediment,the temporal and spatial distribution characteristics and driving factors of the community of denitrifying bacteria were studied.The results showed significant seasonal differences in water parameters and spatial differences in sediment nitrogen(P<0.001).No significant difference(P>0.05)was observed inα-diversity;the EC and SOEF-NH4+-N were important factors affecting theα-diversity of the water and sediment.Denitrifying bacteria mainly belonged to the phylum Proteobacteria and the genera Dechloromonas,Rhodocyclaceae,Pseudomonas,Rhodobacter,and Thauera.Principal coordinate analysis revealed that the community of denitrifying bacteria in the water and sediment exhibited significant spatial differences(P<0.001);keystone denitrifying bacteria in the water also exhibited significant spatial differences(P<0.001).RDA and RF analysis showed that the permanganate index and TP were the main environmental factors affecting the total and keystone denitrifying bacteria in the water;SOEF-NH4+-N,IEF-NH4+-N,and WAEF-NO_(3)--N were the main environmental factors affecting the total and keystone denitrifying bacteria in the sediment.These findings could serve as a reference to understand the interaction mechanism between nitrogen and denitrification bacterial communities in landscape water.
