杂交构树叶多糖发酵条件优化及抗氧化活性研究 被引量:3

Study on antioxidant activity fermentation technology of polysaccharides from hybrid Broussonetia papyrifera leaf

摘要 试验选取酿酒酵母、产朊假丝酵母、扣囊复膜孢酵母为发酵菌种,设计单因素正交试验,研究优化菌种的最优生长条件。以杂交构树叶多糖对自由基(DPPH·)清除率为响应值,通过响应面优化试验,探索其发酵后的抗氧化活性。结果显示,菌种的最优生长条件为:葡萄糖含量6 g/100 mL、蛋白胨的含量1 g/100 mL、生长时间20 h。通过响应面优化试验,最优液态发酵条件:发酵时间60 h、初始pH值4、接种量3%、发酵温度25℃。在此工艺条件下,杂交构树叶多糖DPPH·清除率为78.84%,杂交构树叶多糖发酵后具有较强的抗氧化活性。研究表明,该结果可为杂交构树叶多糖的生物活性研究以及饲用价值开发提供参考。 The experiment selected Saccharom vces cerevisiae, Candida utilis and Saccharomycopsis fibuligera as fermentation strain, and the single factor orthogonal experiment was designed to optimize the optimal growth conditions of the strain. The scavenging rate of free radical(DPPH·) of polysaccharides from Hybrid mulberry leaves was used as the response value, and the antioxidant activity after fermentation was explored through response surface optimization experiment. The results showed that the optimal growth conditions were glucose content 6 g/100 mL, peptone content 1 g/100 mL and growth time 20 h. Through response surface optimization test, the optimal liquid fermentation conditions were fermentation time60 h, initial p H value 4, inoculation amount 3% and fermentation temperature 25 ℃. Under the conditions, the DPPH·clearance rate of hybrid Broussonetia papyrifera leaf polysaccharides was 78.84%, and the hybrid Broussonetia papyrifera leaf polysaccharides had strong antioxidant activity after fermentation. The study indicates that the results can provide a reference for the study of biological activity and feeding value of hybrid Broussonetia papyrifera leaf polysaccharides.
