沙棘叶总黄酮闪式提取工艺优化及组分鉴定 被引量:5

Process Optimization for the Flash Extraction and Component Identification of Total Flavonoids from Seabuckthorn Leaves

摘要 采用闪式法从沙棘叶中提取总黄酮,利用正交试验优化提取工艺条件,并与传统浸提法进行比较。结果表明,闪式法提取沙棘叶总黄酮的最佳工艺为提取电压180 V、提取时间120 s、乙醇体积分数60%、液料比(V_(乙醇)︰m_(沙棘叶)) 18︰1 mL/g。HPLC分析发现粗提液中含有杨梅素、槲皮素、山奈酚和异鼠李素。闪式提取法与传统浸提法提取的沙棘叶总黄酮的得率和粗提物组分基本无差异,但闪式提取短时、低温,能有效降低提取成本。 Total flavonoids were extracted from seabuckthorn leaves by flash extraction method.The extraction conditions were optimized by orthogonal test.The flash extraction of total flavonoids was compared with that of traditional extraction method.The results showed that the optimal flash extraction conditions were extraction voltage 180 V,extraction time 120 s,ethanol volume percentage 60% and liquid-solid ratio(V_(ethanol)︰m_(seabuckthorn leaves)) 18︰1 mL/g.HPLC analysis showed that the flash crude extract contained myricetin,quercetin,kaempferol and isorhamnetin.The flavonoids extraction yield and crude extract flavonoids components of the flash method from seabuckthorn leaves were almost consistent with that of traditional extraction method.However,the short-term and low temperature of flash extraction could effectively reduce the extraction cost.
