复配益生菌对秸秆发酵中总糖含量和活菌数影响的研究

摘要 农作物光合作用一半以上的物质存在于秸秆中。农业废弃物的资源化利用是节能减排和实现可持续发展的重要途径,利用Pseudomonas sp.P11以及其他植物益生菌的复配对千禧果秸秆液态发酵进行了初步探究,首先利用刚果红染色法确定M-12号菌和M-14号菌具有降解纤维素能力,运用改进平板对持法证明纤维素降解菌和P11之间没有明显拮抗作用,第5天测定纤维素酶活和内切葡聚糖酶活力得到M-12号、M-14号菌株活力较P11好,7天总糖含量趋势表明复配菌发酵比单菌发酵效果好,以秸秆发酵液中的有效活菌数确定4种益生菌在一个月内都能不同程度的增殖,其中P11在水溶液中有效活菌数达GB 20287-2006微生物菌肥标准。对于低成本高密度培养P11等益生菌有一定指导作用。 More than half of the photosynthetic substances in crops are found in stalks.The resource utilization of agricultural waste is an important way to save energy,to reduce emissions and to achieve sustainable development.In this paper,a preliminary study was conducted on the liquid fermentation of millennia fruit straw by using the compound of Pseudomonas sp.P11 and other plant probiotics.Firstly,the Congo red dyeing method was used to determine that M-12 and M-14 had the ability to degrade cellulose.There was no obvious antagonistic effect between cellulase degrading bacteria and P11 by using the improved plate pair method.The activity of cellulase and endoglucanase was measured on the 5th day,and the activity of M-12 and M-14 strains was better than that of P11.The trend of total sugar content in the 7th day indicated that the fermentation effect of mixed bacteria was better than that of single bacteria.According to the number of effective viable bacteria in straw fermentation broth,the four kinds of probiotics could all proliferate in different degrees within a month,and the number of effective viable bacteria in water solution of P11 reached the standard of GB 20287-2006.This study has a certain guiding effect on the low-cost and high-density culture of probiotics such as P11.
