威灵仙中锌、锰、铜的初级形态分析及健康风险评价

Primary speciation analysis and health risk assessment of zinc,manganese and copper in Inula nervosa Wall

摘要 为探讨云南威灵仙锌、铜、锰微量元素的形态分布及评价其健康风险,采取正辛醇-水分配体系模拟威灵仙水煎液中锌、铜、锰在人体胃肠中的分配情况。用火焰原子吸收光谱法测定威灵仙原药、水煎液和水溶态、醇溶态中锌、铜、锰的质量比,并对3种微量元素进行健康风险评价。结果表明:云南威灵仙原药中锌、铜和锰元素平均质量比分别为10.10μg/g、21.44μg/g和372.74μg/g,其中锰质量比约为锌质量比的37倍;水煎液锌、铜和锰元素平均质量比分别为3.27μg/g、3.79μg/g和66.47μg/g,溶出率分别为32.42%、17.66%和17.83%,以锌元素的溶出率最高,而铜和锰溶出率较低且基本相同;锌、铜和锰元素主要存在于水溶态中,少量存在于醇溶态中;锌和锰元素的吸收可能分别主要集中在肠部和胃部,铜则在胃、肠中均可能吸收较多;服用原药和水煎液不会对人体产生非致癌健康风险,在醇溶态中3种微量元素也没有非致癌健康风险。从健康风险的角度看,以威灵仙水煎液服用可进一步降低非致癌健康风险;在原药、水煎液和醇溶态中锌、铜、锰对人体的非致癌健康风险程度依次降低。 To investigate the distribution of trace elements of zinc,copper,and manganese in Yunnan Inula nervosa Wall and evaluate its health risks,the n-octanol-water distribution system was adopted to simulate the distribution of zinc,copper,and manganese in the human gastrointestinal tract in the Inala nervosa Wall decoction.The content of zinc,copper,and manganese in the original medicine of Inula nervosa Wall,decoction,water-soluble state and n-octanol-soluble state were determined by flame atomic absorption spectrometry,and health risk assessment of the three trace elements was carried out.The results show that the average content rates of zinc,copper,and manganese in Yunnan Inula nervosa Wall original materials are 10.10μg/g,21.44μg/g,and 372.74μg/g,respectively.The manganese content is about 37 times that of zinc.The average content rates of zinc,copper,and manganese in the decoction are 3.27μg/g,3.79μg/g,and 66.47μg/g,respectively.And the dissolution rates are 32.42%,17.66%,and 17.83%,respectively.The dissolution rate of zinc is the highest,while the dissolution rates of copper and manganese are lower and almost the same.The elements of zinc,copper,and manganese mainly exist in water-soluble state,and a small amount exists in the noctanol-soluble state.The absorption of zinc and manganese may be mainly concentrated in the intestine and stomach,respectively,while copper may be absorbed more in the stomach and intestine.Taking the original medicine and water decoction will not cause non-carcinogenic health risks to the human body,and the three trace elements in the n-octanol-soluble state also have no non-carcinogenic health risks.From the perspective of health risks,taking Inula nervosa Wall decoction can further reduce non-carcinogenic health risks.In the original medicine,decoction and n-octanol-soluble state,the non-carcinogenic health risks of zinc,copper,and manganese to the human body are successively reduced.
