摘要
动脉粥样硬化是大中动脉的慢性炎症性疾病,是众多心血管疾病的病理生理基础。巨噬细胞通过调节脂质浸润和炎症反应参与动脉粥样斑块的形成和发展。髓样细胞触发受体2(TREM2)是一种单次跨膜的免疫球蛋白超家族跨膜受体,能够增强巨噬细胞吞噬能力并抑制其炎性激活,促进巨噬细胞中的胆固醇流出,抑制其向泡沫细胞的转变。泡沫状TREM2型巨噬细胞特异性存在于动脉粥样硬化的粥样斑块中。此外,TREM2脱落后形成的sTREM2亦表现出神经保护和促进炎症反应的功能。因此,本文就巨噬细胞TREM2构造及其信号通路与动脉粥样硬化的相关研究进展进行综述。
Atherosclerosis is a chronic inflammatory disease of large and medium arteries,which is the pathophysiological basis of many cardiovascular diseases.Macrophages are involved in the formation and development of atherosclerotic plaques by regulating lipid infiltration and inflammatory response.Myeloid triggering receptor 2(TREM2)is transmembrane receptors of a single-transmembrane immunoglobulin superfamily that enhance macrophage phagocytosis and inhibit inflammatory activation,promoting cholesterol effluence from macrophages and inhibiting their transformation into foam cells.Foamy macrophages from TREM2 type are specifically present in atherosclerotic plaques.The formation of sTREM2 after the shedding of TREM2 plays a neuroprotective and inflammatory role.In this review,the progress of macrophage TREM2 signaling pathway in atherosclerosis will be discussed.
