摘要
目的研究辛伐他汀辅助治疗精神分裂症的效果及安全性。方法检索了电子数据库(PubMed、Embase、中国生物医学文献数据库和中国知网;检索时间均为建库—2021年5月6日)、相关文献的参考文献、综述以及学会会议上发表的摘要,对评估辛伐他汀辅助治疗精神分裂症的随机对照试验数据进行汇总分析,预先确定的主要结局是阳性和阴性综合征量表(PANSS)评分变化的合并效应量。结果最终纳入了7个研究,包括413例(辛伐他汀辅助治疗组为210例,对照组为203例)。结果显示辛伐他汀组PANSS总分和PANSS一般精神病理量表评分均低于对照组(P<0.05),而PANSS阳性量表评分和阴性量表评分两组比较差异无统计学意义(P>0.05)。结论辛伐他汀辅助治疗可以改善精神症状,尤其是PANSS总分。但鉴于本研究存在的异质性和样本量相对较小,未来将需要更多大样本量的随机对照试验进一步观察不同剂量辛伐他汀对精神分裂症的有效性和安全性。
Objective To explore the efficacy and safety of Simvastatin as adjuvant therapy in treatment of patients with schizophrenia. Methods Electronic databases(PubMed, Embase, Web of Science, CBM and CNKI;retrieval period from database establishment to May 6, 2021), reference documents of pertinent literature, reviews and abstracts presented at related meetings of scientific societies were searched. Data from randomized controlled trials(RCTs) assessing the efficacy of Simvastatin as adjunctive therapy of schizophrenia was collected for pooled analysis. The prespecified primary outcome was the pooled effect size of change in Positive and Negative Syndrome Scale(PANSS) score. Results Finally, 7 studies were included, which included 413 patients(210 patients in Simvastatin as adjuvant therapy group, 203 patients in control group). The results showed that the total PANSS score and general pathological score of PANSS in Simvastatin group were significantly lower than those in control group(P<0.05), but there were no significant differences in positive score and negative score of PANSS between two groups(P>0.05). Conclusion Simvastatin as adjuvant therapy may improve psychiatric symptoms, especially total score of PANSS. However, in view of the significant heterogeneity and the relatively small sample size, more RCTs with larger sample sizes will be needed in the future to further observe the efficacy and safety of different doses of Simvastatin in treatment of patients with schizophrenia.
