百令胶囊对急性胰腺炎模型大鼠肺损伤的修复作用 被引量:2

摘要 目的探讨百令胶囊对急性胰腺炎模型大鼠肺损伤的修复作用。方法将72只Wistar大鼠随机分为假手术组(等体积生理盐水灌胃),模型组(等体积生理盐水灌胃),百令胶囊低、中、高剂量组(1,2,4 g/kg灌胃)及法舒地尔组(30 mg/kg腹腔注射),各12只。除假手术组外,其余5组大鼠均胰胆管注射4%牛磺胆酸钠(1 mL/kg)以复制胰腺炎大鼠模型,建模成功后,各组大鼠均予相应药物或生理盐水,每天1次,连续5 d。采用酶联免疫吸附(ELISA)法测定大鼠血清淀粉酶、白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)及肺组织髓过氧化物酶(MPO)、超氧化物歧化酶(SOD)、活性氧(ROS)水平;采用血气分析仪测定二氧化碳分压(PaCO_(2))、氧分压(PaO_(2)),并计算氧合指数(OI);称定大鼠肺组织湿质量(W)及干质量(D)并计算W/D;采用苏木素-伊红(HE)染色,检查肺组织病理形态学;采用实时定量聚合酶链式反应法及Westernblot法分别测定RhoA、Rho激酶1(ROCK1)、Rho激酶2(ROCK2)mRNA及蛋白表达水平。结果与模型组比较,百令胶囊低、中、高剂量组及法舒地尔组大鼠的血清淀粉酶及IL-6,TNF-α,PaCO_(2),MPO,ROS水平,肺组织W/D及RhoA,ROCK1,ROCK2 mRNA和蛋白水平均显著降低(P<0.05),PaO_(2),OI,SOD水平均显著升高(P<0.05),大鼠肺组织病理形态学显著改善。结论百令胶囊能改善急性胰腺炎肺损伤模型大鼠的肺功能,其机制可能与抑制Rho/ROCK通路,继而抑制炎性因子释放及氧化损伤有关。 Objective To investigate the repairment effect of Bailing Capsules on lung injury in rats with acute pancreatitis.Methods A total of 72 Wistar rats were randomly divided into the sham operation group(gavaged with equal volume of normal saline),model group(gavaged with equal volume of normal saline),Bailing Capsules low-,medium-and high-dose groups(gavaged with 1,2,4 g/kg Bailing Capsules)and fasudil group(intraperitoneally injected with 30 mg/kg,fasudil),with 12 rats in each group.Except for the sham operation group,the rats in the other five groups were injected with 4%sodium taurocholate(1 mg/kg)into the pancreatic duct to replicate the rat model of pancreatitis.After successful modeling,the rats in each group were given corresponding drugs or normal saline once a day for 5 d.The levels of serum amylase,interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)and myeloperoxidase(MPO),superoxide dismutase(SOD)and reactive oxygen species(ROS)in lung tissue were measured by enzyme-linked immunosorbent assay(ELISA)kit.The partial pressure of carbon dioxide(PaCO_(2))and partial pressure of oxygen(PaO_(2))were measured by biochemical analyzer,and the oxygenation index(OI)was calculated.The wet(W)weight and dry(D)weight of rats lung tissue were measured,and W/D weight ratio was calculated.The pathomorphology of lung tissue was examined by hematoxylin-eosin(HE)staining,and RhoA,ROCK1,ROCK2 mRNA and protein expression levels were detected by real-time quantitative polymerase chain reaction(RTq-PCR)and western blot.Results Compared with those in the model group,the levels of serum amylase,IL-6,TNF-α,PaCO_(2),MPO,ROS,lung tissue W/D,the levels of RhoA,ROCK1,ROCK2 mRNA and protein decreased significantly(P<0.05),while the levels of PaO_(2),OI and SOD increased significantly(P<0.05),and the pathomorphology of lung tissue improved significantly in Bailing Capsules low-,medium-and high-dose groups and fasudil group.Conclusion Bailing Capsules can improve the lung function of model rats with lung injury induced by acute pancreatitis,and its mechanism may be related to inhibition of Rho/ROCK pathway and the release of inflammatory factors and oxidative damage.
