番鸭细小病毒YN株分离鉴定及其卵黄抗体效力试验

Isolation and identification of Muscovy duck parvovirus YN strain and its yolk antibody efficacy test

摘要 为了研制预防番鸭细小病毒病的卵黄抗体,试验对采自云南省曲靖市某番鸭养殖场的番鸭细小病毒(Muscovy duck parvovirus, MDPV)阳性病料进行病毒分离和鉴定,用分离株灭活抗原免疫产蛋鸡,制备MDPV卵黄抗体并进行效力试验。结果表明:试验成功分离到MDPV,命名为MDPV YN株;该毒株与GenBank中15株水禽细小病毒的同源性为99.69%~100%,MDPV YN株第626位核苷酸由T突变为C,翻译的209位氨基酸由L变为S。MDPV YN株经番鸭胚传至第26代,病毒纯净、耐氯仿、对1%鸡红细胞无血凝性,检测其病毒含量为1×10^(-5)ELD_(50)/0.2 mL,毒力为1×10^(-5)LD_(50)/0.5 mL。当MDPV卵黄抗体中和抗体效价为1∶256时,用于预防和治疗的剂量分别为0.5 mL/只和2 mL/只。说明MDPV YN株具有良好的免疫原性,以其制备的卵黄抗体对番鸭细小病毒病具有良好的预防和治疗效果。 In order to develop egg yolk antibody for the prevention and treatment of Muscovy Duck parvovirus disease, the experiment was conducted to isolate and identify the virus from a Muscovy duck parvovirus(MDPV) positive diseased material collected from a Muscovy duck farm in Qujing City, Yunnan Province. The inactivated antigen of the isolate was immunized to laying hens for the preparation of MDPV yolk antibody to conduct efficacy test. The results showed that MDPV was isolated successfully and named as MDPV YN strain;the homology between this isolation and 15 isolates of waterfowl parvovirus in GenBank was 99.69%-100%;the 626th nucleotide of MDPV YN strain was changed from T to C, and the translated amino acid 209 was changed from L to S. MDPV YN strain was passed through Muscovy duck embryos to the 26 th generation;the virus was pure, chloroform resistant, and non-hemagglutinating to 1% chicken red blood cells;the virus content was detected to be 1×10^(-5)ELD_(50)/0.2 mL, and the virulence was 1×10^(-5)LD_(50)/0.5 mL. When the MDPV egg yolk neutralizing antibody titer was 1∶256, the doses for prevention and treatment were 0.5 mL/head and 2 mL/head, respectively. The results suggested that MDPV YN strain had good immunogenicity, and its egg yolk antibody had good preventive and therapeutic effect on Muscovy duck parvovirus disease.
