摘要
研究咖啡中4种黄曲霉毒素和杂色曲霉素的快速检测方法并对实际样品进行检测。咖啡样品经酸化的乙腈水溶液超声提取后,通过QuEChERS(quick,easy,cheap,effective,rugged,safe)技术净化,采用超高效液相色谱-串联质谱法进行定性和定量分析。结果表明,5种真菌毒素在1μg/L~20μg/L线性范围内线性关系良好,检出限和定量限分别为0.07μg/kg~0.4μg/kg和0.2μg/kg~1.3μg/kg。4种黄曲霉毒素和杂色曲霉素的平均加标回收率为88.2%~119.0%(n=6),相对标准偏差为1.9%~13.2%。建立的超高效液相色谱-串联质谱法检测方法能够对咖啡豆及咖啡粉样品中黄曲霉毒素和杂色曲霉素进行快速、准确、可靠的分析。
Qualitative and quantitative analyses of aflatoxins(AFB_(1),AFB_(2),AFG_(1) and AFG_(2))and sterigmatocystin(STG)in coffee beans and instant coffee were performed.The samples were ground and extracted with an aqueous solution of acidified acetonitrile.The sample extracts were purified using a“quick,easy,cheap,effective,rugged,safe”(QuEChERS)approach.Aflatoxins and STG were qualitatively and quantitatively analyzed by ultra-high performance liquid chromatography-tandem mass spectrometry.Aflatoxin and STG concentrations had a good linear relationship in the range of 1μg/L-20μg/L.The limit of detectionand limit of quantification were 0.07μg/kg-0.4μg/kg and 0.2μg/kg-1.3μg/kg,respectively.The average recovery of aflatoxins and STG was 88.2%-119.0%(n=6),with relative standard deviations of 1.9%-13.2%.This method is simple and quantitatively accurate and is thus suitable for the rapid analysis of aflatoxins and STG in coffee beans and instant coffee.
