摘要
目的研究覆盆子原花青素对2型糖尿病大鼠糖脂代谢及抗氧化作用的影响。方法用链脲佐菌素(STZ)建立2型糖尿病大鼠模型,将2型糖尿病大鼠随机分为模型组、二甲双胍组、覆盆子原花青素高、中、低剂量组(150 mg/kg、100 mg/kg、50 mg/kg),另设空白组,每组10只。正常组、模型组用等体积的生理盐水灌胃,治疗组按相应剂量每天灌胃,每天1次,连续灌胃8周。观察大鼠的空腹血糖(FPG)、胰岛素(INS)、肝糖原、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)含量,测定谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、丙二醛(MDA)的含量。结果治疗8周后,覆盆子原花青素各剂量组体重较模型组明显升高(P<0.001)。与模型组比较,覆盆子原花青素治疗组FPG明显下降(P<0.01),INS和肝糖原含量增加(P<0.01),TG、TC、LDL-C水平降低、HDL-C水平升高(P<0.05),肝脏GSH-Px、SOD水平明显升高、MDA水平降低(P<0.01,P<0.001)。结论覆盆子原花青素可降低2型糖尿病大鼠的血糖,其作用机制可能与抗氧化、改善糖脂代谢,升高INS和肝糖原含量相关。
Objective To study the effects of raspberry proanthocyanidins on glucose and lipid metabolism and anti-oxidant effects in type 2 diabetic rats.Methods The type 2 diabetic rat model is established with streptozotocin(STZ),and the type 2 diabetic rats are randomly divided into model group,metformin group,raspberry proanthocyanidin high,medium and low dose groups(150 mg/kg,100 mg/kg,50 mg/kg),and a blank group with 10 animals in each group.The normal group and the model group are given an equal volume of normal saline by gavage,and the treatment group is given the corresponding dose by gavage every day,once a day,for 8 weeks.Observe the rats’fasting blood glucose(FPG),insulin(INS),liver glycogen,total cholesterol(TG),triacylglycerol(TC),low-density lipoprotein cholesterol(LDL-C),high-density lipoprotein cholesterol(HDL-C)content,determine the content of glutathione peroxidase(GSH-Px),superoxide dismutase(SOD)and malondialdehyde(MDA).Results After 8 weeks of treatment,the body weight of each dose group of raspberry proanthocyanidin is significantly higher than that of the model group(P<0.001).Compared with the model group,FPG in the raspberry proanthocyanidin treatment group significantly decreased(P<0.01),INS and liver glycogen content increased(P<0.01),TG,TC,LDL-C levels decreased,and HDL-C levels increased(P<0.05),liver GSH-Px,SOD levels are significantly increased,and MDA levels are significantly decreased(P<0.01,P<0.001).Conclusion Raspberry proanthocyanidins can reduce blood sugar in type 2 diabetic rats,and its mechanism of action may be related to anti-oxidant,improvement of glucose and lipid metabolism,and increase of INS and liver glycogen content.
