摘要
【目的】培育适合广西种植的抗螟虫转基因杂交水稻恢复系,为广西杂交水稻抗虫育种提供基础材料。【方法】以广西杂交稻骨干恢复系亲本广恢998、桂恢1561和桂恢110为受体,以携带Bt抗虫基因cry1C*的抗螟虫转基因材料T1C-19为供体,通过1次杂交、3次回交和8次自交,利用分子标记辅助选择(MAS)技术,结合抗虫鉴定(室内离体接虫鉴定与室外自然鉴定)、苏云金芽孢杆菌(Baclillus thuringiensis)蛋白(Bt蛋白)含量测定和农艺性状选择培育适合广西推广应用的抗螟虫杂交水稻恢复系。【结果】对经过杂交、回交和自交育成的3个抗螟虫转基因恢复系KH998、KH1561和KH110进行农艺性状考查,结果表明,KH998、KH1561和KH110的千粒重均低于供体亲本T1C-19,KH1561的千粒重比原始亲本桂恢1561低,而KH998和KH110的千粒重较原始亲本分别提高1.40和3.40 g,产量优势变大;以KH110组配的杂交组合天丰A/KH110的产量比对照天优华占(CK2)增加1.29%,但所组配的其他杂交组合产量均低于对照特优63(CK1)和CK2。抗虫鉴定结果表明,KH998、KH1561和KH110及其配配杂交组合均具有与抗虫供体亲本T1C-19相当的抗性,均可有效抵御螟虫危害;杀虫蛋白含量测定结果表明,KH998、KH1561和KH110的平均Bt蛋白含量分别为0.24、0.65和1.41μg/g FW,其中,KH998和KH1561抗性群体的Bt蛋白能稳定表达,而KH110抗性群体的Bt蛋白表达不够稳定。【结论】通过回交转育结合MAS育成的3个抗螟虫转基因恢复系KH998、KH1561和KH110均可稳定遗传cry1C*基因,并成功表达Bt杀虫蛋白,且具有与供体亲本T1C-19相当的抗螟虫能力。
[Objective]Cultivating the restorer line which is resistant to borers and is suitable for planting in Guangxican can provide material basis for the breeding of hybrid rice with resistance to borers in Guangxi.[Method]The backbone restorer lines of Guanghui 998,Guihui 1561 and Guihui 110 were used as receptors and the stem borer resistant transgenic material T1C-19 carrying Bt insect resistant gene cryl C^(*)was used as donors to cultivate restorer lines of hybrid rice with stem borer resistance.One hybridization,three backcrosses and eight selfcrossings were conducted combined with molecular marker assisted selection,insect resistance identification(indoor isolated insect docking i-dentification and outdoor natural identification),Baclillus thuringiensis protein(Bt protein)content determination and agronomic character selection.[Result]Three insect resistant restorer lines KH998,KH1561 and KH110 were tested for agronomic characters.Thousand-grain weight of KH998,KH1561 and KH110 were lower than the donor parent T1C-19,and that of KH1561 was lower than the original parent Guihui 1561,while those of KH998 and KH110 increased by 1.40 and 3.40 g,respectively with a increasing yield advantage.The KH110 hybrid Tianfeng A/KH110 increased by 1.29%than the control Tianyouhua(CK2),but the other crosses were lower than the control Teyou 63(CK1)and CK2.The insect identification showed that KH998,KH1561 and KH110 and their mating crosses had resistance equivalent to insect-resistant parent T1C-19,and all resistant to borer.The average Bt protein contents of KH998,KH1561 and KH110 were 0.24,0.65 and 1.41μg/g FW,respectively.Bt protein in KH998 and KH1561 resistant populations was stable,but not in KH110 resistant population.[Conclusion]The three insect resistant restorer lines KH998,KH1561 and KH110 bred by backcross transfer and molecular marker assisted selection can stably inherit cryl C^(*)gene,successfully express Bt insecticidal protein,and have the same insect resistance as donor parent T1C-19.
