摘要
目的探讨麻黄附子细辛汤对阳虚型非小细胞肺癌小鼠PI3K/Akt/mTOR信号通路的影响。方法C57BL/6小鼠按随机分配分为模型组,化疗组,麻黄附子细辛汤高、中、低剂量组(中药高、中、低组),联合组(麻黄附子细辛汤高剂量组+顺铂),建立阳虚型非小细胞肺癌小鼠模型。造模成功后进行分组给药,采用Western Blot法检测小鼠肿瘤组织PI3K/Akt/mTOR通路相关蛋白表达水平。结果①麻黄附子细辛汤、化疗药可减弱PI3K p85a、PI3K p110a蛋白表达水平,暂可认为两药存在交互协同作用;②麻黄附子细辛汤与化疗药物合用对降低Akt、p-Akt蛋白表达具有较明显的协同作用;③麻黄附子细辛汤和顺铂联用对减弱mTOR、p-mTOR蛋白的表达具有协同作用。结论本实验从整体药效方面探讨麻黄附子细辛汤阻断阳虚型非小细胞肺癌小鼠PI3K/Akt/mTOR信号通路的机制,研究发现麻黄附子细辛汤可一定程度上减弱PI3K、Akt、mTOR蛋白的表达水平,对PI3K/Akt/mTOR信号通路的传导具备一定的抑制功效。
Objective To investigate the effect of Mahuang Fuzi Xixin decoction on PI3K/Akt/mTOR signal pathway in Yang deficiency non-small cell lung cancer mice.Methods C57BL/6 mice were randomly divided into model group,chemotherapy group,Mahuang Fuzi Xixin Decoction high,medium and low dose group(traditional Chinese medicine high,medium and low group),combined group(Mahuang Fuzi Xixin Decoction high dose group+cisplatin).After the successful establishment of the model,the drugs were administered in groups,and the expression level of PI3K/Akt/mTOR pathway related proteins in mouse tumor tissues was detected by Western blot.Results 1.Mahuang Fuzi Xixin decoction and chemotherapeutic drugs can reduce the protein expression levels of PI3K p85a and PI3K p110a.It can be considered that there is interaction and synergy between the two drugs for the time being;2.Mahuang Fuzi Xixin decoction combined with chemotherapy drugs has obvious synergistic effect on reducing the expression of Akt and p-Akt protein;3.The combination of Mahuang Fuzi Xixin decoction and cisplatin has a synergistic effect on weakening the expression of mTOR and p-mTOR proteins.Conclusion This experiment discusses the mechanism of Mahuang Fuzi Xixin decoction blocking PI3K/Akt/mTOR signal pathway in Yang deficiency non-small cell lung cancer mice from the aspect of overall efficacy.It is found that Mahuang Fuzi Xixin decoction can reduce the expression level of PI3K,Akt and mTOR protein to a certain extent,and has a certain inhibitory effect on the transmission of PI3K/Akt/mTOR signal pathway.
