摘要
为探讨PHB对颈总动脉阻塞诱导的短暂脑缺血小鼠模型中缺血再灌注损伤的神经保护作用,将C57/BL6小鼠随机分为正常组、模型组和模型+PHB组。结果表明:脑内PHB过表达可显著缓解脑缺血小鼠海马区神经元的变性、坏死和凋亡,提高模型组小鼠脑内的血流量。PHB可通过抑制脑缺血小鼠脑内的炎症反应和上调自噬功能发挥神经保护作用。PHB还可通过促进脑缺血小鼠脑内的神经元再生起到神经保护作用。综合而言,PHB可通过促进脑血流量、神经元再生、抑制脑内神经炎症反应、上调脑内自噬反应等起到神经保护作用,为治疗人类脑梗塞及其他神经系统疾病提供了重要的依据。
To explore the vital role of PHB on ischemia-reperfusion injury in mice with transient cerebral ischemia established by common cerebral artery occlusion.Intracerebral injection of AAV9-PHB significantly alleviated the necrosis and apoptosis of neurons in hippocampus of cerebral ischemia mice,and can increase the cerebral blood flow in cerebral ischemia mice.In addition,PHB can significantly inhibit the neuroinflammation in cerebral ischemia mice,and activate the autophagy inside the cerebral ischemia mice.Further study showed that PHB could promote neuronal regeneration.In summary,intracerebral injection of AAV9-PHB can play a neuroprotective role by promoting cerebral blood flow and neuronal regeneration,inhibiting neuroinflammatory reaction in brain and promoting autophagy reaction inside brain.This study provides an effective biological target protein for the treatment of cerebral ischemia injury and other neurological disease.
