摘要
【目的】比较2种不同配方高脂饲料构建高脂血症合并脂肪肝大鼠模型的差异。【方法】将36只雄性SD大鼠随机分为对照组、模型I组和模型Ⅱ组,分别给予维持饲料、配方I高脂饲料和配方Ⅱ高脂饲料,在0、1、5、7周时测定血生化指标,终期取肝脏称质量,采用苏木素-伊红(HE)染色法观察肝组织病理学表现。【结果】模型Ⅰ组大鼠体质量持续增加,实验结束时,血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)水平均显著升高,高密度脂蛋白胆固醇(HDL-C)水平显著降低,谷丙转氨酶(ALT)、谷草转氨酶(AST)水平升高,肝脏指数明显增加,肝脏脂肪变达到重度水平。模型Ⅱ组大鼠体质量无明显升高,血清中TC、LDL-C水平均升高,HDL-C水平明显降低,ALT、AST水平升高,肝脏指数明显增加,肝脏脂肪变达到中度水平。【结论】配方Ⅰ高脂饲料可建立混合型高脂血症合并脂肪肝大鼠模型,配方Ⅱ高脂饲料可用于建立高胆固醇症合并脂肪肝大鼠模型的研究。
Objective To compare the difference of two formulations of high-fat diets in constructing hyperlipidemia(HLP)combined with fatty liver rat model.Methods Thirty-six male SD rats were randomly divided into the control group,model groupⅠand model groupⅡ,by feeding maintenance diet,Formula I highfat diet and FormulaⅡhigh-fat diet,respectively.The serum biochemical indices were determined at 0,1,5 and 7 weeks.At the final stage,the liver was weighed and the histopathological findings were observed by hematoxylin-eosin(HE)staining.Results The body mass of rats in the model groupⅠwas increased continuously,and at the end of the experiment,the serum levels of total cholesterol(TC),triglycerides(TG)and low-density lipoprotein cholesterol(LDL-C)were significantly increased,high-density lipoprotein cholesterol(HDL-C)level was significantly decreased,alanine aminotransferase(ALT)and aspartate transaminase(AST)levels were increased,liver index was significantly increased,and liver steatosis reached a severe level.In the model groupⅡ,there was no significant increase in body mass,while the serum TC and LDL-C levels were increased,HDL-C level was significantly decreased,ALT and AST levels were increased,liver index was significantly increased,and liver steatosis changes arrived to the moderate level.Conclusion The formulaⅠhighfat diet can be used to establish the model of mixed HLP complicated with fatty liver,and formulaⅡhigh-fat diet can be used to establish the model of high cholesterol complicated with fatty liver.
