摘要
为完善芍药种丛生芽诱导途径,以芍药种胚为外植体,研究接种方式(传统切割、只留胚切割、半保留胚乳切割)对种胚启动培养的影响,基本培养基(1/2MS、MS、WPM)、去除子叶、预处理(暗培养、含活性炭(AC)培养基预转接)、外源激素(6-BA、TDZ、GA_(3))及浓度(0.3,0.6,1.0mg·L^(-1))对丛生芽增殖的影响,以及AC(0,1.0,2.0,3.0,4.0g·L^(-1))、IAA和IBA组合(0,0.5,1.0mg·L^(-1))对丛生苗生根的影响。结果表明:保留胚乳越少,胚萌发速度越快,半保留胚乳接种的种胚萌发效果最优;切除胚苗子叶诱导丛生芽速度快诱导率高,最佳培养基为WPM+1.0mg·L^(-1)GA_(3)+1.0mg·L^(-1)6-BA;腋芽增殖培养前在3.0g·L^(-1)AC培养基上进行5d的暗培养,能够有效降低转接后的褐化率;最适宜腋芽生长的6-BA浓度为0.6mg·L^(-1),GA_(3)浓度为0.3mg·L^(-1);低浓度生长素诱导生根效果不佳,诱导丛生苗生根最佳培养基为1/2MS+1.0mg·L^(-1)IBA+3.0g·L^(-1)AC。
In order to improve the pathway for the induction of multiple shoots from Paeonia lactiflora seeds,Paeonia lactiflora embryos were used as explants to study the effects of inoculation methods(traditional cutting,embryo-reserved cutting and semi preserved endosperm cutting)on the initiation culture of embryos.The effects of basic medium(1/2MS,MS and WPM),cotyledon removal,pretreatment(dark culture,pre-transfer with AC medium),exogenous hormones(6-BA,TDZ and GA_(3)),and concentrations(0.3,0.6,1.0mg·L^(-1))on the proliferation of multiple shoots were investigated.And the effects of AC(0,1.0,2.0,3.0,4.0g·L^(-1)),IAA and IBA combinations(0,0.5,1.0mg·L^(-1))on the rooting of clustered seedlings.The results showed that the less the retained endosperm was,the faster the embryo germination was.The embryo germination effect of semi-preserved endosperm inoculation was the best.The rapid induction rate of multiple shoots was high after cotyledons were excised,and the optimal medium was WPM+1.0mg·L^(-1)GA_(3)+1.0mg·L^(-1)6-BA.Five day dark culture on 3.0g·L^(-1)AC medium before axillary bud proliferation culture could effectively reduce the browning rate after transfer.The optimal concentration of 6-BA and GA_(3) for axillary bud growth was 0.6mg·L^(-1)and 0.3mg·L^(-1),respectively.The rooting effect of low concentration auxin was not good,and the optimal medium for inducing the rooting of clustered seedlings was 1/2MS+1.0mg·L^(-1)IBA+3.0g·L^(-1)AC.
