摘要
为了探寻对松果梢斑螟有高致病性的病原微生物,本研究分离培养了采自秦皇岛北戴河区的感染松果梢斑螟幼虫病原物,结合菌种形态特征及ITS序列分析鉴定明确了其主要致病菌种,测定了其致病力。结果表明,该菌株为球孢白僵菌(编号Gq-Bb菌株),当孢子悬浮液浓度从1×10^(4)孢子/mL逐步提高到1×10^(8)孢子/mL时,其对各虫龄松果梢斑螟幼虫的致病性逐渐增强。当浓度为1×10^(8)孢子/mL时,其1龄和2龄幼虫的累计校正死亡率均达到100%,3龄、4龄和5龄幼虫的分别为96%、90%和77.33%。1~5龄幼虫的LC_(50)值分别为1.26×10^(3)、1.90×10^(3)、2.99×10^(3)、2.12×10^(4)、2.12×10^(5)孢子/mL,LT_(50)分别为4.34~2.57、4.91~2.78、5.19~2.78、5.73~2.99和6.44~3.64 d,结果表明随该菌孢子悬浮液浓度提高,其致病力逐渐增强;在孢子悬浮液浓度为1×10^(8)孢子/mL时,其对1~4龄松果梢斑螟幼虫具有较高致病力。该结果为松果梢斑螟的生物防治提供新的菌株资源和参考。
In order to isolate the pathogenic microorganisms that are highly pathogenic to Dioryctria pryeri,the pathogenic microorganisms were cultured and separated from the Dioryctria pryeri larvae sampled from Beidaihe District,Qinhuangdao.The main pathogenic bacteria were identified by combined analysis of the morphological characteristics and ITS sequences and their pathogenicity were subsequently determined.A pathogenic strain was identified as Beauveria bassiana(named Gq-Bb),whose pathogenicity to larvae of various instars gradually increased as the spore concentration increased from 1×10^(4)to 1×10^(8)spores/mL.When the concentration was 1×10^(8)spores/mL,the cumulative corrected mortality of the 1^(st) and 2^(nd) instar larvae reached 100%,while the mortality of the 3^(rd),4^(th) and 5^(th) instar larvae were 96%,90%and 77.33%.The LC_(50) values of 1^(st)-5^(th) instar larvae were 1.26×10^(3),1.90×10^(3),2.99×10^(3),2.12×10^(4),2.12×10^(5)spores/mL,and their LT_(50) were 4.34 to 2.57 d,4.91 to 2.78 d,5.19 to 2.78 d,5.73-2.99 d and 6.44-3.64 d.It showed that the virulence gradually increased as the concentration of the spore suspension of the fungus increased,and the highest virulence to the larvae of Dioryctria pryeri from 1^(st) instars to 4^(th) instars appeared when the spore suspension concentration was 1×10^(8)spores/mL.This result provides a new strain resource and reference for the biological control of Dioryctria pryeri.
