摘要
探讨和血生络方对缺血性卒中大鼠神经功能的影响及其作用机制。方法成年SD雄性大鼠随机分为Sham组、MCAO组、HX组和HX+k252a组,采用大脑中动脉阻断法诱导建立大鼠脑缺血模型(Sham组除外),HX组和HX+k252a组灌胃给予和血生络方药液,每日1次,持续14d,Sham组和MCAO组给予等量生理盐水灌胃。采用TTC染色法测定脑缺血大鼠的脑梗死体积;采用网屏实验评价大鼠的前肢肌力和Ludmila Belayev 12分评分法评价大鼠感觉运动整合能力。免疫荧光染色观察BrdU+/Nestin+细胞在侧脑室室下区(SVZ)的表达;qRT-PCR法检测SVZ中Nestin、BDNF及TrkB的基因表达;Western Blot检测SVZ中Nestin、BDNF及TrkB的蛋白表达。结果与MCAO组比较,在PMD3和PMD7,HX组大鼠脑梗死体积显著减少(P<0.05),在PMD7和PMD14,HX组大鼠的前肢肌力显著增强(P<0.05),PMD3、PMD7和PMD14,HX组大鼠的感觉运动整合能力显著增强;SVZ中BrdU+/Nestin+细胞表达显著增多(P<0.05),Nestin、BDNF、TrkB的蛋白和基因表达均显著增加(P<0.05)。与HX+k252a组比较,HX组大鼠脑梗死体积减小无显著性差异(P>0.05),HX组大鼠的前肢肌力和感觉运动整合能力显著增强(P<0.05);SVZ中BrdU+/Nestin+细胞表达显著增多(P<0.05),Nestin、BDNF、TrkB的蛋白和基因表达均显著增加(P<0.05)。结论和血生络方能够改善脑缺血大鼠的神经功能,这可能与上调BDNF-TrkB信号通路的表达,促进SVZ神经干细胞增殖有关。
Objective To observe effects of HXSLF on neurological function and its related mechanisms in MCAO rats.Methods The middle cerebral artery occlusion method was used to induce cerebral ischemia model in rats(except the sham group).HX group and HX+k252 a group were administered HXSLF decoction by gavage once a day for 14 days.Sham group and vehicle group were received equal volume of saline.The volume of cerebral infarction was measured by TTC staining,and the neurological function was evaluated by screen test and Ludmila belayev 12 score.The BrdU+/nestin+cells in the subventricular zone(SVZ)was observed by immunofluorescence staining;the mRNA expression of nestin,BDNF and TrkB in SVZ was measured by qRT-PCR;the protein expression of nestin,BDNF and TrkB in SVZ was detected by Western blot.Results Compared with MCAO group,in HX group,the infarct volume of MCAO rats was significantly decreased on PMD3 and PMD 7(P<0.05),forepaws strength of MCAO rats were enhanced significantly(P<0.05),on PMD3,PMD7 and PMD14 sensory motor integration ability of MCAO rats was grown up significantly(P<0.05),BrdU+/nestin+cell expression in SVZ was risen significantly(P<0.05),protein and mRNA expression of nestin,BDNF and TrkB were increased significantly(P<0.05).Compared with HX+k252 a group,in HX group,there was no significant difference in the volume of infarction(P>0.05),forepaws strength of MCAO rats were stronger(P<0.05),on PMD3,PMD7 and PMD14 sensory motor integration ability of MCAO rats was enhanced significantly(P<0.05),BrdU+/nestin+cell expression in SVZ was risen significantly(P<0.05),protein and mRNA expression of nestin,BDNF and TrkB were stimulated significantly(P<0.05).Conclusion HXSLF could significantly improve the neurological functional deficit of MCAO rats,which may be related to the up regulation of BDNF-TrkB signal pathway and the proliferation of neural stem cells in SVZ.
