仙人掌果花色苷纯化工艺的研究 被引量:2

摘要 仙人掌果中含花色苷、黄酮等多种具有生物活性的物质,其中花色苷具有抗氧化、预防慢性疾病、抑制炎症,改善视力、调节肠道菌群等作用。为了对仙人掌果中花色苷进行深入研究,本文主要对其纯化工艺进行优化,选用HPD-100树脂,静态吸附与解析条件为:静态吸附时间为6h,上样液浓度为90mg/L,上样液pH2,静态解析时间为4h,解析液浓度为80%;动态吸附与解析条件为:柱床体积90mL,动态吸附流速为2mL/min,动态解析流速为1mL/min,除去可溶性杂质用水量为100mL。纯化后花色苷的色价是纯化之前9.33倍(P<0.05),纯化后花色苷的纯度是纯化之前的5.44倍(P<0.05)。通过高效液相色谱进行定量分析,得到仙人掌果粉中花色苷含量为806.4mg/100g(以矢车菊素-3-O-葡萄糖苷计)。 Opuntia ficus-indica contains anthocyanins,flavonoids and other substances rich in biological activity.Among them,anthocyanins have the functions of anti-oxidation,preventing chronic diseases,inhibiting inflammation,improving eyesight,and regulating intestinal flora.In order to conduct an in-depth study on the anthocyanins,the crude anthocyanin solution was purified in this experiment,and HPD-100 resin was selected for purification.The static adsorption and analysis conditions were as follows:static adsorption time was 6h,and the concentration of the sample solution was 90mg/L,the pH of the sample solution was 2,the static analysis time was 4h,and the concentration of the analysis solution was 80%.The dynamic adsorption and analysis conditions were as follows:the column bed volume was 90mL,the dynamic adsorption flow rate was 2mL/min,and the dynamic analysis flow rate was 1mL/min,the amount of water used to remove soluble impurities was 100mL.After purification,the color value of anthocyanins increased significantly(P<0.05),which was 9.33 times before purification,and the purity of anthocyanins after purification was significantly improved(P<0.05),which was 5.44 times before purification.And the content of anthocyanin in opuntia ficus-indica powder was 806.4mg/100g(calculated as Cyanidin-3-glucoside)by high-performance liquid chromatography quantitative analysis.
