摘要
目的评价葵肽清风对高尿酸血症动物模型尿酸水平的影响,并探讨其可能的作用机制。方法77只SPF级Wistar大鼠随机分为7组,分别为正常对照组、模型组、秋水仙碱组、苯溴马隆组及葵肽清风低、中、高浓度组,模型组制备大鼠高尿酸血症和急性痛风性关节炎复合模型,造模成功后,各给药组给予相应的药物干预,测定大鼠血尿酸浓度、黄嘌呤氧化酶、关节肿胀程度。结果中药干预各组血尿酸水平较模型组均显著降低(P<0.05),其中以高浓度组下降最显著,呈剂量依赖性;中药干预各组黄嘌呤氧化酶活性均有显著降低,并且其中高浓度组下降最显著;中药高浓度组在48 h关节肿胀程度与12、24 h相比显著减小(P<0.01),且中药高浓度组在48 h关节直径已恢复正常,作用优于秋水仙碱组和苯溴马隆组。结论葵肽清风能在控制血尿酸水平和痛风局部症状两个层次上治疗急性痛风性关节炎。
Objective To evaluate the effect of Kuitai Qingfeng Drinks on the uric acid level in an animal model of hyperuricemia,and to explore its possible mechanism of action.Methods A total of seventy-seven SPF Wistar rats were randomly divided into 7 groups:a normal control group,a model group,a colchicine group,a benzbromarone group and Kuitai Qingfeng Drinks low-,medium-and high-concentration groups.The compound model of hyperuricemia and acute gouty arthritis was established.After that,each drug-administered group was intervened by giving the corresponding drugs,and the levels of blood uric acid concentration,xanthine oxidase,and joint swelling degree in the rats were measured.Results The level of serum uric acid in each group treated with traditional Chinese medicine was significantly lower than that in the model group(P<0.05),among which the high-concentration group was most significantly decreased,in a dose-dependent manner.The XOD activity was decreased significantly in each of the three Kuitai Qingfeng Drinks groups,and the decrease was the most significant in the high-concentration group.The degree of joint swelling in the high-concentration group was decreased significantly at the 48th hour compared with that at the 12th or 24th hour(P<0.01),and its joint diameter returned to normal at the 48th hour,whose effect was better than that in the colchicine group and the benzbromarone group.Conclusion Kuitai Qingfeng Drinks can treat acute gouty arthritis by controlling the level of blood uric acid and improving local symptoms of gout.
