基于网络药理学方法探讨玉屏风散治疗慢性阻塞性肺疾病的分子机制研究 被引量:2

摘要 目的观察基于网络药理学方法分析慢性阻塞性肺疾病予以玉屏风散治疗的分子机制研究。方法利用网络药理学数据库筛选玉屏风散的有效成分,选择由广州中医药大学动物实验中心提供的健康SD大鼠70只,分为模型组、正常对照组,玉屏风低、中、高剂量组,泼尼松组、泼尼松联合玉屏风中剂量组,每组10只,Western Blot检测肺组织中水通道蛋白5(AQP5)、黏蛋白5(MUC5AC)的表达;RT-PCR法检测肺组织中AQP5、MUC5AC mRNA的表达。结果除正常对照组外,泼尼松联合玉屏风中剂量组的AQP5蛋白(0.59±0.16)、其mRNA(2.48±0.19)明显高于其他各组,差异有统计学意义(P<0.05);玉屏风散组随着剂量的增加,AQP5蛋白、其mRNA逐渐增高(P<0.05);除正常对照组外,泼尼松联合玉屏风中剂量组的MUC5AC蛋白(0.27±0.05)、其mRNA(1.25±0.03)指标明显低于其他各组,差异有统计学意义(P<0.05);玉屏风散组随着剂量的增加,MUC5AC蛋白、mRNA含量逐渐减低,差异有统计学意义(P<0.05)。结论慢性阻塞性肺疾病进行玉屏风散治疗,可促使肺组织中的AQP5蛋白增高,而MUC5AC蛋白水平下降,纠正黏蛋白/水盐比例紊乱是其治疗COPD的分子机制。 Objective To observe and analyze the molecular mechanism of Yupingfengsan for the treatment of chronic obstructive pulmonary disease based on network pharmacology.Methods The effective ingredients of Yupingfeng Powder were screened by the network pharmacology database,and 70 healthy SD rats provided by the Animal Experiment Center of Guangzhou University of Traditional Chinese Medicine were selected.Divided into model group,normal control group,Yupingfeng low,medium and high dose group,prednisone group,prednisone combined with Yupingfeng medium dose group,each with 10 animals.Western Blot detects the expression of aquaporin 5(AQP5)and mucin 5(MUC5AC)in lung tissue.RT-PCR method was used to detect the expression of AQP5 and MUC5AC mRNA in lung tissue.Results Except for the normal control group,the AQP5 protein(0.59±0.16)and mRNA(2.48±0.19)of the prednisone combined with Yupingfeng medium-dose group were significantly higher than those of the other groups,and the difference was statistically significant(P<0.05).With the increase of the dose in the Yupingfengsan group,AQP5 protein and mRNA gradually increased,and the difference was statistically significant(P<0.05).Except for the normal control group,the MUC5AC protein(0.27±0.05)and mRNA(1.25±0.03)indexes of the prednisone combined with Yupingfeng medium-dose group were significantly lower than those of the other groups,and the difference was statistically significant(P<0.05).With the increase of the dose in the Yupingfengsan group,the protein and mRNA content of MUC5AC gradually decreased,and the difference was statistically significant(P<0.05).Conclusion Yupingfeng powder treatment for chronic obstructive pulmonary disease can increase the AQP5 protein in the lung tissue,while the MUC5AC protein level decreased.Correcting the disorder of the mucin/water-salt ratio was its molecular mechanism for the treatment of COPD.
