响应曲面法优化西藏黑青稞中多酚提取工艺

Optimization of Polyphenol Extraction Technology from Black Tibetan Hulless Barley by Response Surface Methodology

摘要 在比较5个青稞品种籽粒中多酚含量的差异后,选取相对含量高、产自西藏山南隆子县的黑青稞为试验本体,为优化其所含多酚物质的提取工艺,在单因素试验的基础上,根据响应面法,获得最适的工艺优化条件为:提取温度50℃,提取时间70min,乙醇体积分数60%,料液比(g︰mL)1︰20,从而得到提取量为3.03 mg/g的多酚物质。 The present study determined optimal conditions for extraction of polyphenols from black Tibetan hulless barley.First,five varieties of Tibetan hulless barley were compared for polyphenol content;as a result,the black hulless barley produced in Longzi County,Shannan,Tibet was found to have the highest relative content of polyphenols,so it was selected as the experimental material for optimization of polyphenol extraction technology.The study of optimization of this technology was conducted on the basis of a single factor test,using response surface method.Consequently,the optimum extraction conditions were identified as follows:the extraction temperature was at 50℃,the extraction time was 70 min,the volume fraction of ethanol was 60%,and the ratio of solid to solvent was 1∶20(g∶mL).Under these conditions,the average rate of polyphenol extraction reached 3.03±0.51 mg/g.
