摘要
目的 探讨同一物种的细胞(同样的糖基酶体)在体外和体内表达的蛋白质糖基化表达谱的差异,为体内和体外细胞培养蛋白质糖基化提供实验依据。方法 选取人结肠癌细胞系HCT116、RKO、DLD和10例结肠癌患者的血清为实验对象,分别代表体外培养与体内合成,进行蛋白质糖基化表达水平分析,比较分析同一物种的细胞在体外和体内表达的蛋白质糖基化表达谱的差异。结果 结肠癌患者血清蛋白质N-糖基化表达中高甘露糖型较少,多为双天线复合型糖,不含平分型,与结肠癌细胞HCT116存在一定相似性,而与结肠癌细胞RKO、DLD差异较大。结论 体外培养细胞的蛋白质糖基化,与体内细胞的蛋白质糖基化表达并不完全一致。因此在蛋白质糖基化相关的研究中,体内验证是必不可少的一个环节,需要引起生物标志物、药物或疫苗研究领域的重视。
Objective To investigate the differences in protein glycosylation expression profiles of cells of the same species(same glycosylase) expressed in vitro and in vivo, and to provide experimental evidence for protein glycosylation between in vivo and in vitro cell culture. Method Human colon cancer cell lines HCT116, RKO, DLD, and serum from10 colon cancer patients were selected as the experimental objects, representing in vitro culture and in vivo synthesis, respectively. The protein glycosylation expression level analysis was performed to compare and analyze the differences in the protein glycosylation expression profiles expressed in vitro and in vivo by cells of the same species. Result There were fewer high-mannose types in the expression of serum protein N-glycosylation, most of which were biantennary complex glycoform, without bisecting types In patients with colorectal cancer. The expression type in patients was similar to HCT116 cells, however, quite different from RKO and DLD cells. Conclusion The novel glycosylation type of proteins in cells cultured in vitro is not completely consistent with the expression of protein glycosylation in cells in vivo. Therefore, in vivo validation is an indispensable point in the research related to protein glycosylation, which requires attention in the field of biomarker, drug, or vaccine research.
