丁酸梭菌和枯草芽孢杆菌对高邮鸭盲肠内容物短链脂肪酸含量及盲肠组织转录组影响研究

Effects of Clostridium butyricum and Bacillus subtilis on Short Chain Fatty Acids in Cecal Contents and Transcriptome of Cecal Tissues in Gaoyou Ducks

摘要 试验旨在研究饲粮中添加丁酸梭菌和枯草芽孢杆菌对育雏育成期高邮鸭盲肠内短链脂肪酸含量影响以及盲肠组织转录调控作用。选择1日龄健康、体重相近的高邮公鸭90只,随机分3组,每组3个重复,每个重复10只。对照组饲喂基础日粮,丁酸梭菌组添加丁酸梭菌活菌3×10;cfu/g,枯草芽孢杆菌组添加枯草芽孢杆菌活菌5×10;cfu/g,试验期60 d。结果显示:饲粮中添加丁酸梭菌显著增加盲肠内容物中乙酸、丙酸和丁酸浓度(P<0.05);盲肠组织转录测序分析发现,与对照组相比,丁酸梭菌添加组和枯草芽孢杆菌添加组盲肠组织分别分析到299和382个差异表达基因,而丁酸梭菌添加组和枯草芽孢杆菌添加组比较,两者盲肠组织中仅11个差异表达基因;差异表达基因聚类分析结果发现,与对照组相比,添加组差异基因富集的多个通路与跨膜转运、跨膜转运蛋白活性(GO富集分析)以及细胞外基质受体的相互作用(KEGG富集分析)相关。综上所述,饲粮中添加丁酸梭菌显著提高盲肠内容物中乙酸、丙酸和丁酸浓度,添加丁酸梭菌和枯草芽孢杆菌影响盲肠组织转录表达,且两种不同添加菌对盲肠组织转录调控表现出一定程度的一致性。 This experiment was performed to evaluate the effects of dietary supplementation of Clostridium butyricum and Bacillus subtilis on the cecal concentration of short chain fatty acids and the transcriptional regulation of cecal tissue in Gaoyou ducks during the breeding period. A total of 90 healthy 1-day-age Gaoyou male ducks with average body weight were randomly divided into 3 groups with 3 replicates(10 ducks per replicate). The control group was fed basal diet, and the Clostridium butyricum and Bacillus subtilis groups were fed basal diet added with live Clostridium butyricum(3×10;cfu/g) and Bacillus subtilis(5×10;cfu/g), respectively. The experiment lasted for 60 days. The results showed that Clostridium butyricum supplementation significantly increased the concentrations of acetic acid, propionic acid and butyric acid in cecal contents(P<0.05). Transcriptional sequencing analysis of cecal tissue showed that 299 and 382 genes were differentially expressed in Clostridium butyricum and Bacillus subtilis groups when compared with the control group, respectively. And only 11 genes were differentially expressed between Clostridium butyricum and Bacillus subtilis groups. Function enrichment analysis showed that several GO pathways were associated with transmembrane transport, transmembrane transporter activity, and interaction of extracellular matrix receptor was detected by KEGG analysis. In conclusion, dietary Clostridium butyricum affected the concentrations of short chain fatty acids including acetic acid,propionic acid and butyric acid in cecal content, the addition of Clostridium butyricum and Bacillus subtilis in duck diet affected the transcriptional expression of cecal tissue similarly to a certain exlent.
