摘要
目的探讨银翘散对急性肺损伤(ALI)模型小鼠肺损伤的改善及作用机制。方法将60只KM小鼠随机分为阴性对照组、模型组、银翘散组、脱氧核糖核酸酶Ⅰ(DNaseⅠ)组,各15只。银翘散组灌胃给予银翘散(按生药和体质量计72.54 g/kg),阴性对照组、模型组、DnaseⅠ组小鼠均灌胃给予等体积生理盐水,每天1次,连续3 d。末次给药30 min后,腹腔注射10%水合氯醛麻醉小鼠,剥离皮下组织,暴露气管,于气管内滴注脂多糖(5 mg/kg),阴性对照组同法滴注生理盐水,以复制ALI模型小鼠;DNaseⅠ组建模成功10 min后同法滴注DNaseⅠ(5 mg/kg)。6 h后处死小鼠,取出肺组织,测定肺部湿/干重比(W/D);采用免疫荧光法和蛋白免疫印迹法检测髓过氧化物酶(MPO)和瓜氨酸化组蛋白H3(CitH3)的表达水平;苏木精-伊红(HE)染色,观察肺组织的病理形态。结果与模型组比较,银翘散组W/D显著降低(P<0.05),MPO和CitH3的表达水平均显著降低(P<0.05),气道周围及肺组织中的炎性细胞浸润减轻。结论银翘散可通过抑制中性粒细胞胞外诱捕网形成中MPO和CitH3的表达水平,从而减轻小鼠ALI。
Objective To investigate the improvement and mechanism of Yinqiao Powder on the lung injury in acute lung injury(ALI)model mice.Methods A total of 60 KM mice were randomly divided into the negative control group,model group,Yinqiao Powder group and deoxyribonucleaseⅠ(DNaseⅠ)group,with 15 mice in each group.Mice in the Yinqiao Powder group were gavaged with Yinqiao Powder(calculated by the crude drug and body mass,72.54 g/kg),while mice in the negative control group,model group and DNaseⅠgroup were gavaged with equal volume of normal saline,once a day for 3 d.The mice were anesthetized by intraperitoneal injection of 10%chloral hydrate 30 min after the last administration,the subcutaneous tissue was stripped,the trachea was exposed,lipopolysaccharide(5 mg/kg)was injected into the trachea,and mice in the negative control group were injected with normal saline in the same way to replicate the ALI model mice.Mice in the DNaseⅠgroup were injected with DNaseⅠ(5 mg/kg)in the same way 10 min after the successful modeling.The mice were killed after 6 h,the lung tissue was taken out,and the wet-to-dry weight ratio(W/D)of the lung was measured.Immunofluorescence and Western blot methods were used to detect the expression levels of myeloperoxidase(MPO)and citrullinated histone H3(CitH3).Hematoxylin-eosin(HE)staining was used to observe the pathological morphology of lung tissue.Results Compared with those in the model group,the W/D in Yinqiao Powder group decreased significantly(P<0.05),the expression levels of MPO and CitH3 in Yinqiao Powder group decreased significantly(P<0.05),and the infiltration of inflammatory cells around the airway and lung tissue decreased.Conclusion Yinqiao Powder can reduce the ALI in mice by inhibiting the expression levels of MPO and CitH3 in the formation of neutrophil extracellular traps.
