光化学衍生法测定玉米及玉米制品中的黄曲霉毒素B_(1) 被引量:1

摘要 为研究玉米及玉米制品被黄曲霉毒素B_(1)污染的情况,确保市场上玉米及玉米制品的食用安全,本文旨在建立免疫亲和柱净化-高效液相色谱-柱后光化学衍生-荧光法测定玉米和玉米制品中黄曲霉毒素B_(1)含量的方法。将玉米和玉米制品试样粉碎后,试样粉末在甲醇-水溶液(体积比为55:45)条件下提取,经免疫亲和柱富集和净化后,以甲醇-水溶液(体积比45:55)为流动相,采用XDB-C18色谱柱(5μm×4.6 mm×250 mm),由光化学柱后衍生,荧光检测器检测(Ex:360 nm,Em:440 nm)。结果表明:黄曲霉毒素B_(1)的最低检出限和定量检出限分别为0.009μg/kg和0.03μg/kg,标准曲线的线性范围为0.1~0.5 mg/L(R=0.9990),2个不同水平的加标平均回收率为88.4%~90.1%,RSD值为0.47%~1.07%。该方法具有操作简便、灵敏度高、重现性佳等特点,适用于玉米和玉米制品中黄曲霉毒素B_(1)的检测。 In order to investigate the contamination status of aflatoxin B_(1) in corn and corn products and ensure the food safety of corn and corn products in the market,a method was established to determine the content of aflatoxin B_(1) in corn and corn products by immunoaffinity column purification and HPLC-FLD coupled with post-column photochemical derivatization.After the samples of corn and corn products were made into powder,aflatoxin B_(1) in corn and corn products powder was extracted with methanol-water solution(volume ratio 55:45),and purified with immunoaffinity column.Aflatoxin B_(1) was separated by HPLC with post-column photochemical derivatization in which methanol-water solution(volume ratio 45:55)was used as the mobile phase and XDB-C18 column(5μm×4.6 mm×250 mm)was used.Then it was quantified with fluorescence detector(Ex:360 nm,Em:440 nm).The results showed that the minimum and quantitative detection limit of aflatoxin B_(1) were 0.009μg/kg and 0.03μg/kg respectively,and the linear range of the standard curve was 0.1~0.5 mg/L(R=0.9990).The average recovery rate of different levels of standard addition was 88.4%~90.1%,and the RSD value was 0.47%~1.07%.This method was suitable for the detection of aflatoxin B_(1) in corn and corn products because of simple operation,high sensitivity and good reproducibility.
