摘要
目的:探讨调控miR-140-3p表达对非小细胞肺癌(non-small cell lung carcinoma,NSCLC)细胞株A549和NCI-H1299细胞行为的影响。方法:体外培养A549和NCI-H1299细胞,转染miR-140-3p mimic(过表达)或miR-140-3p inhibitor(抑制表达),通过EdU检测细胞增殖率,通过Transwell及划痕实验检测细胞迁移能力,通过生物信息学工具预测miR-140-3p的潜在靶基因,通过生物信息学软件构建基因调控网络。结果:过表达miR-140-3p后,A549和NCI-H1299细胞的增殖率升高,迁移能力下降;相反,抑制miR-140-3p的表达后,细胞的增殖率较低,但迁移能力提高。生物信息学分析结合实时定量反转录聚合酶链式反应验证提示,miR-140-3p可能靶向CBL基因,影响细胞生物学过程。结论:miR-140-3p对NSCLC细胞具有促进细胞增殖和抑制细胞迁移的作用。
Objective:To investigate the effect of regulating the expression of miR-140-3p on the behavior of non-small cell lung cancer(NSCLC)cell lines A549 and NCI-H1299.Methods:A549 and NCI-H1299 cells were transfected with miR-140-3p mimic(overexpression)or miR-140-3p inhibitor(inhibition)in vitro,respectively.Cell proliferation rate was detected by EdU,and cell migration ability was detected by Transwell and wound healing assay.The potential target genes of miR-140-3p were predicted by bioinformatics tools,and the gene regulatory network was constructed by bioinformatics software.Results:A549 and NCI-H1299 cells transfected with miR-140-3p mimic exhibited increased proliferation rate and decreased migration ability.On the contrast,A549 and NCI-H1299 cells transfected with miR-140-3p inhibitor had lower proliferation rate but higher migration ability.Bioinformatic analysis and quantitative real-time reverse transcription polymerase chain reaction results revealed that miR-140-3p might target CBL and mediate many biological processes.Conclusion:miR-140-3p can promote the cell proliferation and inhibit cell migration of NSCLC cells.
