摘要
目的探讨龙贝逍遥散冻干粉(Longbei Xiaoyao Powder,LXP)联合阿霉素(Adriamycin,ADM)对乳腺癌耐阿霉素细胞株MDA-MB-231/ADR细胞的增殖与迁移作用及可能作用机制。方法MDA-MB-231/ADR细胞中分别加入DMSO、ADM(1μg/ml)、LXP(0.8 mg/ml)+ADM、自噬增强剂雷帕霉素(rapamycin,RA)(1μg/ml)+LXP+ADM培养液1ml,CCK-8法及细胞划痕实验检测其对各组细胞增殖与迁移影响,qPCR检测实验各组MDA-MB-231/ADR细胞耐药基因MDR1、MRP1,及自噬调节基因Rubicon的mRNA表达。结果LXP能增强ADM对MDA-MB-231/ADR的细胞增殖抑制作用(P<0.01,P<0.001),并能增强ADM对MDA-MB-231/ADR细胞迁移抑制作用(P<0.01)。ADM联合LXP后,能降低MDA-MB-231/ADR细胞MDR1及MRP1基因表达(P<0.05),加入自噬增强剂RA后,LXP的这一作用被逆转。LXP组+ADM组MDA-MB-231/ADR细胞的Rubicon表达较DMSO组及ADM组均明显上升(P<0.01),当加入自噬增强剂RA后,LXP这一作用被逆转,表明LXP能抑制MDA-MB-231/ADR细胞自噬。结论LXP可能通过调节MDA-MB-231/ADR细胞自噬,降低MDA-MB-231/ADR细胞多药耐药基因表达,进而逆转MDA-MB-231/ADR细胞多药耐药。
Objective To investigate the effects of Longbei Xiaoyao Powder(LXP)combined with Adriamycin(ADM)on the proliferation and migration of breast cancer adriamycin-resistant cell line MDA-MB-231/ADR cells mechanism.Methods MDA-MB-231/ADR cells were added with DMSO,ADM(1μg/ml),LXP(0.8 mg/ml)+ADM,and rapamycin,an autophagy enhancer,The proliferation and migration of MDA-MB-231/ADR cells were detected by CCK8 method and cell scratch test.The mRNA expression of MDR1,MRP1,and Rubicon in MDA-MB-231/ADR cells were detected by qPCR.Results LXP could enhance the inhibitory effect of ADM on MDA-MB-231/ADR cell proliferation(P<0.01,P<0.001),and could enhance the inhibitory effect of ADM on MDA-MB-231/ADR cell migration(P<0.01).ADM combined with LXP could reduce the expression of MDR1 and MRP1 genes in MDA-MB-231/ADR cells(P<0.05),and this effect of LXP was reversed by adding the autophagy enhancer RA.The expression of Rubicon in MDA-MB-231/ADR cells in LXP group+ADM group was significantly higher than that in DMSO group and ADM group(P<0.01).When the autophagy enhancer RA was added,the effect of LXP was reversed,indicating that LXP can Inhibits autophagy in MDA-MB-231/ADR cells.Conclusion LXP may reverse the multidrug resistance of MDA-MB-231/ADR cells by regulating autophagy and reducing the expression of multidrug resistance genes in MDA-MB-231/ADR cells.
