运动与电针对肥胖大鼠海马内炎症因子的影响

Effects of Treadmill Exercise and Electroacupuncture on Inflammatory Factors in the Hippocampus of Obese Rats

摘要 目的:探讨4周有氧耐力运动与电针单用或运动联合电针对肥胖大鼠海马内炎症因子的影响。方法:将SD大鼠随机分为普通饲料(C)组与高脂饲料组(H)。8周后,随机选取C组8只大鼠为对照组(NC)。选取H组大鼠为实验组,并随机分为肥胖安静(OC)、肥胖运动(OE)、肥胖电针(OA)与肥胖运动+电针组(OEA)。NC、OC组不进行运动;OE组大鼠经1周适应性运动后,进行4周中等强度的运动;OA组大鼠经1周适应性针刺后,进行4周正式电针干预;OEA组大鼠进行4周运动和电针的联合干预。实验结束后,各组大鼠禁食过夜,10%水合氯醛麻醉,冰上分离海马,ELISA法检测海马组织内IL-6、IL-10、MCP-1、IL-1β和TNF-a的表达。结果:(1)电针引起肥胖大鼠海马内IL-10含量显著升高;(2)运动联合电针导致肥胖大鼠海马内IL-1β含量显著升高,IL-10和IL-6含量出现显著降低。结论:实验证实单用电针或运动联合电针干预对肥胖大鼠海马内相关炎症因子产生了显著性影响,且运动联合电针的干预效果更加显著。 Objective:To discuss about the impact of 4-week of aerobic endurance treadmill exercise training and electroacupuncture alone or both on inflammatory factors in the hippocampus of obese rats.Methods:5-week-old male SD rats were randomly divided into normal diet group(C)and high fat diet group(H).After eight weeks,Eight rats in Group C were randomly selected as normal diet sedentary group(NC),and rats in group H,whose body weight values were above the mean value plus 1.4 times the standard deviations of the body weight of rats in group H were picked out as obese rats and then randomly divided into obese sedentary group(OC),obesity exercise group(OE),obese electroacupuncture group(OA)and obesity exercise combined with electroacupuncture group(OEA).Rats in group NC and group OC were without treadmill exercise training.Rats in group OE were performed 4-week moderate intensity treadmill exercise training after being adapted to the treadmill for a week.Rats in group OA were carried out 4-week electroacupuncture after adapting to electroacupuncture for a week,and rats in the OEA group were performed a combination of 4-week treadmill exercise training and electroacupuncture.All rats were fasted overnight at the end of the experiment,anesthetized with 10%chloral hydrate.Hippocampus were isolated in ice bath.The expressions of IL-6,IL-10,MCP-1,IL-1βand TNF-α in the hippocampus tissues were detected by ELISA.Results:(1)The content of IL-10 in the hippocampus tissues in obese rats was significantly increased by electroacupuncture;(2)The IL-1βcontent in the hippocampus tissues of obese rats was significantly increased by exercise combined with electroacupuncture,and significantly decreased in IL-10 and IL-6 contents.Conclusion:The experiment demonstrated that electroacupuncture or exercise training combined with electroacupuncture had a significant impact on inflammation factors in the hippocampus of obese rats,and especially the impact of exercise training combined with electroacupuncture on that was more significant.
