摘要
目的研究白芍总苷对排卵障碍大鼠排卵功能及性激素水平的影响及相关机制。方法建立排卵障碍大鼠模型,随机分为排卵障碍组、抑制剂组、白芍总苷组和抑制剂+白芍总苷组,另设健康组。抑制剂组以SB4315425 mg·kg^(-1)腹腔注射,白芍总苷组以白芍总苷200 mg·kg^(-1)灌胃,抑制剂+白芍总苷组用SB431542+白芍总苷干预。检测血清性激素、抗米勒管激素(anti-Müllerian hormone,AMH)水平;HE染色观察卵巢形态学变化,计数单侧卵巢中成熟卵泡数、生长卵泡数、黄体数;用Western blot检测卵巢组织转化生长因子β1(transforming growth factorβ1,TGF-β1)、Smad4、Smad3、p-Smad3蛋白的表达水平。结果抑制剂组、排卵障碍组、抑制剂+白芍总苷组、白芍总苷组血清促黄体素(lutropin,LH)、卵泡刺激素(follicle-stimulating hormone,FSH)、雌二醇(estradiol,E2)、AMH水平依次升高,成熟卵泡数、生长卵泡数、黄体数依次增加(P<0.05)。HE染色显示,排卵障碍组中黄体及差异发育卵泡的数量明显减少,可观察到较多囊状扩张卵泡,卵泡中颗粒细胞层数消失或明显减少,抑制剂组的变化更为明显;抑制剂+白芍总苷组、白芍总苷组始基卵泡、初级卵泡及黄体的数量增加,白芍总苷组的改善更为明显。抑制剂组、排卵障碍组、抑制剂+白芍总苷组、白芍总苷组TGF-β1、Smad4、Smad3、p-Smad3蛋白的相对表达量依次升高(P<0.05)。结论白芍总苷可提高排卵障碍大鼠性激素、AMH水平,改善排卵功能、卵巢组织形态学变化,可能通过激活TGF-β1/Smads通路来实现。
Objective To study the effects of total glucosides of paeony on ovulation function and sex hormone levels in rats with ovulation disorders,and to explore the related mechanisms.Methods Rat models of ovulation disorder were constructed and randomly divided into ovulation disorder group,inhibitor group,paeony total glycosides group,inhibitor+paeony total glycosides group,and another healthy group.The inhibitor group were injected with SB4315425 mg·kg^(-1) intraperitoneally,the total glucosides group were administered intragastrically with total glucosides of paeony 200 mg·kg^(-1),and the paeony total glucosides group were treated with SB431542 and total glucosides of paeony.The serum sex hormones and anti-Mullerian hormone(AMH)levels were detected.The morphological changes of the ovary were observed by HE staining and the numbers of mature follicles,growing follicles and corpus luteum in unilateral ovaries were counted.Western blot was used to detect the expression of transforming growth factor β1(TGF-β1),Smad4,Smad3,and p-Smad3 in ovarian tissue.Results The serum luteinizing hormone(LH),follicle-stimulating hormone(FSH),estradiol(E2),and AMH levels,the number of mature follicles,the number of growing follicles,and the number of corpus luteum in the inhibitor group,ovulation disorder group,inhibitor+total glucosides of paeony group,and total glucosides of paeony group were increased sequentially(P<0.05).HE staining showed that the corpus luteum and differentially developed follicles were significantly reduced in the ovulation disorder group,and more cystic expanded follicles were observed.The number of granulosa cell layers in the follicles disappeared or decreased significantly,and the changes in the inhibitor group were more obvious.The number of primordial follicles,primary follicles and corpus luteum increased in the total glycosides group and the total glucosides of paeony group,and the improvement was more obvious in the total glucosides group.The relative expression of TGF-β1,Smad4,Smad3,and p-Smad3 protein in the inhibitor group,ovulation disorder group,inhibitor+total glucosides of paeony and total glucosides of paeony were increased sequentially(P<0.05).Conclusion The total glucosides of paeony can increase the levels of sex hormones and AMH in rats with ovulation disorders,improve the ovulation function and morphological changes of ovaries,which may be achieved by activating the TGF-β1/Smads pathway.
